人参糖蛋白的结构及其镇静安眠作用研究 被引量:13

摘要 从人参水提取物部分分离得到糖蛋白组分,此糖蛋白中性糖含量为37.27%、蛋白含量为53.98%、酸性糖含量为10.45%。高效液相色谱法测定重均分子量为17 731u的混合物。糖链部分由6种单糖组成,鼠李糖(Rha)、甘露糖(Man)、葡萄糖(Glc)与半乳糖(Gal)为非还原末端,其中以Glc为主,而Rha仅以非还原末端形式存在于糖链中;主链以(1→2)与(1→4)-连接的半乳糖醛酸(Gal A)和(1→4)-连接的Glc构成,其中主要为(1→4)-连接的Glc,同时存在少量分支结构,即(1→3,4)、(1→4,6)与(1→3,6)-连接的甘露糖。-消除反应测定结果表明,人参糖蛋白含有O-糖肽键,且N-乙酰氨基半乳糖胺(Gal NAc)是通过(1→4)-连接与氨基酸成键;蛋白部分由17种氨基酸组成。人参糖蛋白腹腔注射20mg/kg时,可显著降低小鼠自主活动次数(P<0.05);可显著减少阈下剂量戊巴比妥钠所致小鼠的睡眠潜伏期,并延长睡眠时间(P<0.05)。人参糖蛋白具有镇静安眠的作用。 Study on the structure and sedative hypnotic effects of the glycoprotein, which was obtained from water extract of Panaxginseng. Its neutral sugar content was 37.27%, protein content was 53.98%, and acid sugar content was 10.45%. And it was a mixture which weight average molecular weight was 17731 u analyzed by HPLC and GPC software. The sugar chains were consist of 6 kinds of monosaccharide, non reducing ends were Rha,Man,Glc and Gal, which was mainly Glc, and Rha only exist in the form of non reducing ends in the sugar chain. The backbone were composed of (1→2) and (1→4)-linked GalA, and (1→4)-linked Glc, which was mainly (1 →4)-linked Glc, and there were also a small amount of branches which were (1 →3,4), (1→4, 6) and (1→3,6)-linked Man. The result of 13-elimination reaction showed it was O-linked glycoprotein. The GalNAc connected to amino acid by (1 →4)-linkage. The ginseng gly- coprotein (i.p., at the dose of 20mg/kg) could significantly reduce the number of activities in mice, and had synergistic effect on pento- barbital sodium of under threshold dose, can significantly reduced the sleep latency (P 〈 0.05), prolong the sleeping time of mice. So the ginseng glycoprotein showed sedative hypnotic effects.
