摘要
目的:通过实时无标记细胞分析法与传统MTT法对乳腺癌细胞增殖进行监测以对比确定研究细胞增殖的最佳实验方法手段。方法:采用MTT法、实时无标记细胞分析法测定瘦素刺激乳腺癌MCF-7细胞增殖的效果。结果:MTT法和实时无标记细胞分析法结果均表明瘦素作用浓度为0.625 nmol/L、作用时间为24 h的条件下对乳腺癌MCF-7细胞有诱导生长的效果。但实时无标记细胞分析法能够实时监测细胞生长和增殖的动态生物学反应过程,观察效果更加直观、准确。结论:实时无标记细胞分析法可较好地弥补MTT法的缺点,成为一种新的研究细胞增殖的实验方法。
Objective: To detect the proliferation of breast cancer MCF-7 cell by real time cell analysis (RTCA) and traditional MTT method to identify the better methods. Method: MTI" and RTCA methods were used to monitor the proliferation of breast cancer cell MCF-7 induced by different concentrations of Leptin (0-62.5 nmol/L). Results: MTT and RTCA all showed that Leptin with the concentration of 0.625 nmol/L and 24 h of the incubation could get the optimal effect for MCF-7 cell proliferation. But the result of RTCA was more intuitive and accurate. Conclusion: RTCA may be a better way to offset the shortcomings of the MTT assay, and it will become an applied means for cell proliferation.
