抑郁症外周血circRNA的生物信息学分析 被引量:5

Bioinformatic analysis of peripheral blood circRNA in depression disorder

摘要 目的采用生物信息学方法预测hsa_circRNA_102802和hsa_circRNA_104597相对应的miRNA的靶基因,并分析其所参与的生物学过程及信号通路。方法应用targetscan,miranda和mirbase预测上述2个circRNA对应的miRNA的靶基因,各个miRNA的预测结果分别取交集后,再取其合集,应用FunNet进行功能富集分析和信号通路富集分析。结果用3个在线数据库得到hsa_circRNA_102802对应的miRNA(has_miR_204_5p,has_miR_809,has_miR_520_5p,has_miR_423_5p,hasmiR_617,has_miR_877_5p)的靶基因的合集为199个,hsa_circRNA_104597对应的miRNA(has_miR_659_3p,has_miR_9_5p,has_miR_661,has_miR_576_3p,has_miR548d_5p,has_miR_548a_5p,has_miR_548b_5p,has_miR_8765p,has_miR_744_5p)的靶基因的合集为410个;靶基因所涉及的生物学过程广泛,包括多项与中枢神经系统相关的条目,如皮层发育、轴突导向和延伸、突触传递、以及学习和记忆过程等(尸〈0.05);所涉及的信号通路包括轴突导向、谷氨酸能突触、Wnt信号通路、ErbB信号通路、mTOR信号通路、VEGF信号通路等与抑郁症存在密切联系;hsa_circRNA_102802和hsa_cir_cRNA_104597在抑郁症中可能起着更加重要的作用。结论hsa_circRNA_102802和hsa_circRNA_104597可能与抑郁症的发病机制密切相关。 Objective To predict the target genes of miRNA associated with hsa circRNA_102802 and hsa_circRNA_l(M597 using bioinformatics methods, and analyse the biological process and signaling pathway. Methods Targetscan, miranda and mirbase three online database were used to predict target genes of miRNAs which are complementary to circRNAs. Target genes of miRNAs prediction results were taken their intersection of three online database, then take their collection as total target genes of miRNAs. The re- suits were analyzed by gene ontology(GO) and KEGG pathway analysis using FunNet. Results 199 target genes of has miR 2(M_Sp, has_miR_809, has_miR 520 5p, has_miR_423_Sp, has_miR_617 and has_miR 877_5p were gotten from the intersection of 3 databases; and 410 targets of has_miR_659_3p, has_miR_9 5p,has_miR_661,has miR 576_3p,has_miR_548d_5p,has_miR_548a5p,has_miR_548b_5p,has_miR 876_5p and has_miR_744_Sp were gotten from the intersection of 3 databases. GO analysis showed that Tar- get genes involved in diverse biological processes including central nervous system, such as cortex develop- ment, axon guidance and extension, synaptic transmission, as well as the process of learning and memory (P〈 0.05). Enriched pathways were also revealed target genes involved in the signal path including the axon guidance,glutamic acid synapses, Wnt signaling pathway, ErbB signaling pathway, mTOR signaling pathway and VEGF signaling pathway, which connect closeiy with depression, hsa_circRNA_102802 and hsa_cir- cRNA_104597 may play an important role in depression.Conclusions hsa_circRNA_102802 and hsa_cir-cRNA_104597 may be associated with the pathologenesis of depression disorder.
