摘要
为建立蒙古栎扦插繁殖技术体系,探究其嫩枝扦插的生根机理。以当年生蒙古栎中上部嫩枝条做插穗,采用ABT 1号、IBA、NAA及IBA+NAA混合溶液处理,分别采取200、300、500mg·L^(-1)与1 000mg·L^(-1)浓度处理不同的时间,以清水处理为对照。采用高效液相色谱法对其生根过程中吲哚乙酸(IAA)、脱落酸(ABA)、玉米素(ZT)、赤霉酸(GA3)4种内源激素含量进行测定。结果表明,用生长调节剂IBA 1 000mg·L^(-1)处理10s和IBA+NAA 1 000mg·L^(-1)处理10s可有效促进愈伤组织发育,但生根率仅为3.3%。在生根过程中IAA与ZT均可有效促进愈伤组织而促进生根,ABA抑制生根,低浓度GA3有利于不定根的形成。插穗的愈伤组织发生能力、生根能力与IAA/ABA比值和ZT/ABA比值呈正相关。
In order cuttings of Querc Q. mongolica wer NAA, and mixed to establish an efficient cutting propagation system, the rooting mechanism of softwood was explored. The current year shoots in the middle and upper branches of tings,which were treated by different growth regulators of ABT No. 1, IBA, with gradient concentrations of 200,300,500 mg · L-1 ,and 1 000 mg · L -1 for different time durations, and water was used as the control. Four endogenous hormones, i. e. , indoleace- tic acid (IAA), abscisic acid (ABA), zeatin (ZT), and gibberellin (GA3) during rooting were determined by high performance liquid chromatography (HPLC). The results showed that the treatment with IBA 1 000 mg · L-1 for 10 s and IBA+NAA 1 000 mg · L- 1 for 10 s could promote the callus development,but the rooting rate was only 3.3 %. In the process of rooting, IAA and ZT could promote the development of callus and promote the rooting. ABA inhibited the rooting,while low level of GA3 was beneficial to adventitious root formation. There was a positive correlation between IAA/ABA ratio and ZT/ABA ratio in callus growth and rooting ability.
