摘要 目的探讨CUEDC2(CUE domain containing 2)在不同侵袭、迁移能力结直肠癌细胞系中的表达。方法培养结直肠癌细胞系SW480、SW620和LOVO,Transwell小室实验检测结直肠癌细胞系的侵袭、迁移能力,RealTime PCR与Western Blot分别检测CUEDC2的mRNA及蛋白水平的表达。结果结直肠癌细胞系SW480、SW620、LOVO侵袭、迁移能力逐渐增强,差异具有统计学意义(均P<0.05)。结直肠癌细胞系SW480、SW620、LOVO表达CUEDC2mRNA依次增加,LOVO、SW620表达量明显高于SW480,差异具有统计学意义(P<0.05)。结直肠癌细胞系SW480、SW620、LOVO的CUEDC2蛋白表达量依次增加,LOVO、SW620表达量明显高于SW480,差异具有统计学意义(P<0.05)。结论随着结直肠癌细胞侵袭迁移能力的增加,CUEDC2的表达水平具有增加的趋势。CUEDC2可能具有促进结直肠癌细胞系侵袭迁移的作用,有望成为潜在的预测结直肠癌侵袭转移的标识蛋白。 Objective To investigate the expression of CUE domain containing 2(CUEDC2)in colorectal cancer cell lines with different invasion and migration ability.Methods The invasion and migration ability of colorectal cancer cell lines SW480,SW620 and LOVO was detected by transwell chamber assay.Real-time PCR and Western blot were used to detect the expression of CUEDC2 mRNA and protein,respectively.Results The invasion and migration ability and CUEDC2 mRNA and protein expression increased orderly in SW480,SW620 and LOVO cells(P<0.05).Compared with SW480 cells,the expression of CUEDC2 mRNA and protein significantly increased in SW620 and LOVO cells(P<0.05).Conclusion The expression of CUEDC2 shows an increasing trend with the increased invasion and migration ability in colorectal cancer cells.CUEDC2 may play an important role in promoting the invasion and migration of colorectal cancer cells and may be a potential marker for predicting the invasion and metastasis of colorectal cancer.
