摘要
目的以沙苑子为原料,研究简单、高效的提取、分离、纯化沙苑子苷A的方法。方法以沙苑子苷A的含量为指标,采用正交试验法优化醇提工艺,所得浸膏经萃取、聚酰胺、硅胶柱分离,甲醇重结晶纯化沙苑子苷A,用波谱法、薄层色谱(TLC)法、高效液相色谱(HPLC)法检查纯度。结果最佳提取工艺条件为,提取次数为3次,加20倍乙醇,提取时间为90 min,乙醇体积分数为70%;分离纯化的沙苑子苷A经波谱法、TLC法检查与对照品一致,经HPLC法检测纯度超过98.50%。结论优选出的最佳提取工艺提取率高、稳定性好,可为沙苑子苷A的工业化生产工艺提供指导。分离工艺已达到中药质量检测用化学对照品技术要求,所选用的分离、纯化方法简单、安全,可行性强,成本低且效率高。
Objective To study a simple and efficient method of extracting,separating and purifying of Complanatoside A from Astragalus Complanatus. Methods The extraction process was optimized by orthogonal test. The extract was separated by extraction,polyamide and silica gel column,and Complanatoside A was purified by methanol recrystallization. The purity was checked by spectral,TLC,HPLC methods. Results The optimum extraction conditions were as follows:the extraction time was 3 times,the ratio of liquid to material was 1 :20,the extraction time was 90 min,and the ethanol concentration was 70%. The separation and purification of Complanatoside A ac-corded with the reference was tested by spectral TLC method. The purity was more than 98. 50% tested by HPLC method. Conclusion The best extraction process has the advantages of high extraction rate and good stability,which can provide guidance for the industrial pro-duction process of Complanatoside A. The separation process has reached the technical requirements of chemical reference materials for tra-ditional Chinese medicine quality detection. The separation and purification method is simple,safe,feasible,low cost and high efficiency.
