莲子心总碱毒性和对四氯化碳急性肝损伤的保护作用 被引量:6

摘要 目的探讨莲子心总生物碱毒性及对四氯化碳急性肝损伤的保护作用。方法一次性给予给药组昆明小鼠(10只)口服灌胃量2000 mg/kg莲子心总碱,对照组昆明小鼠(10只)给予同等剂量0.3%羧甲基纤维素钠,密切观察72 h内两组小鼠的表现中毒症状以评价其急性毒性。SD大鼠随机分为6组,分别作为空白对照组,模型组,阳性对照组(水飞蓟素200 mg/kg),莲子心总生物碱低、中、高剂量(100、200、400 mg/kg)组,以四氯化碳为诱导剂进行肝损伤诱导,造模24 h后再禁食16 h,检测大鼠血清中丙氨酸氨基转移酶(ALT)、碱性磷酸酶(ALP)、总胆红素(TBIL)、天冬氨酸氨基转移酶(AST)水平,肝组织匀浆液中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化酶(GSH-Px)以及丙二醛(MDA)活性,同时测定肝脏系数,并观察肝脏组织病理切片。结果对于昆明小鼠莲子心总生物碱安全剂量可高达2000 mg/(kg·d)。100、200、400 mg/kg莲子心总生物碱可改善模型组大鼠肝脏系数,逆转受损大鼠血清中ALT、AST、ALP以及TBIL水平(P<0.05),恢复受损肝组织中氧化酶SOD、CAT、GSH-Px的含量,降低MDA水平(P<0.05),各指标变换趋势与剂量呈相关性。结论莲子心总生物碱具有较高的安全性以及对急性肝损伤具有保护作用。 Objective To discuss toxicity of Lotus plumule and its hepatoprotective for carbon tetrachloride leading a- cute hepatic damage. Methods Kunming mice (n=lO) in drug group received 2000 mg/kg TALP, while Kunming mice in control group (n=10) received 0.3% sodium carboxymethyl cellulose at the same dose, then poisoning symptoms of mice were closely observed in 72 h and acute toxicity was evaluated. SD rats were randomly divided into 6 groups: blank control group, model group, positive control group (silymarin 200 mg/kg), Lotus root total alkaloids low, medium and high dose group (100, 200, 400 mg/kg), carbon tetrachloride induced acute liver injury, after molding 24 h and fasted for 16 h, the levels of serum alanine aminotransferase (ALT), alkaline phosphatase (ALP), total bilirubin (TBIL) and aspartate aminotrans ferase (AST) in serum,and the content of superoxide dismutase (SOD), catalase (CAT), glu- tathione over activity of peroxidase (GSH-PX) and the content of malondialdehyde (MDA) were determined, the liver organ coefficient was also measured and pathological sections of the liver were observed. Results For Kunming mice, Lotus total alkaloid safe dose was as high as 2000 mg/(kg.d). Liver organs coefficient of rats could be improved with i00, 200 and 400 mg/kg Lotus total alkaloid. The ALT, AST, ALP and TBIL in serum of injured rats were reversed (P 〈 0.05), the SOD, CAT, GSH-Px recovered, and MDA contents reduced (P 〈 0.05), changes tendency of index was relat- ed with dosage. Conclusion Lotus total alkaloid has high safety and protective effect on acute liver injury.
