摘要
目的:探讨PI3K抑制剂联合吡柔比星对人膀胱癌BIU-87细胞增殖作用的影响。方法:用不同浓度的PI3K抑制剂LY294002联合吡柔比星对BIU-87细胞作用0、12、24、36 h后,通过MTT方法检测抑瘤率;通过Western blotting检测Caspase-3、Bax、Bcl-2及P-gp蛋白的表达。结果:THP能明显抑制BIU-87细胞的增殖,并且具有明显的时间的依赖性(P<0.05)。THP联合应用不同浓度LY294002组与单独使用同剂量的THP组比较,BIU-87细胞的增殖随着药物浓度的增加和时间的延长进一步受到抑制,而且,联合应用LY294002的低浓度THP组对BIU-87细胞抑制作用明显强于高剂量THP组(P<0.05)。此外,PI3K抑制剂LY294002联合应用THP后,BIU-87细胞Bax与Caspase-3表达都明显上升(P<0.05),Bcl-2与P-gp表达量明显降低(P<0.05)。结论:PI3K抑制剂联合THP比单独应用THP对人膀胱癌BIU-87细胞抑制作用效果明显。
ObjectiveTo investigate the effects of PI3K inhibitor LY294002 combined with Pirarubicin （T H P） on proliferationof human bladder cancer cells BIU-87. Methods：The BIU-87 cells were treated by the different concentration of LY294002 and THP for0 ,1 2 ,2 4 ,3 6 h. The tumor inhibition rate was measured with MTT. The Bax, B cl-2, Caspase-3 and P-gp of BIU-87 cells were detected byWestern blotting. Results：THP could significantly inhibit the proliferation of BIU-87 cells and had a significant time dependence （ P 〈0 .0 5 ）. Compared the THP combined with different concentration of LY294002 groups and used alone at the same dose in THP group,the proliferation of BIU-87 cells increased with the increase of drug concentration and time further inhibited, and the low concentrationTHP group combined with LY294002 on the inhibition of BIU-87 cells was stronger than the high dose THP group （P 〈 0. 0 5 ）. Furthermore,the expressions of Bax and Caspae-3 in cytoplasm of BIU-87 cells treated by LY294002 and THP were increasing more than thecontrol group（ P 〈 0. 05） , while the expressions of Bcl-2 and P-gp were decreasing（ P 〈 0. 0 5 ）. Conclusion ： It is better inhibiting effecton BIU-87 cell to use LY294002 and THP combined treatment than only THP.
