摘要
【目的】探明粳型优势生态型水稻中各品种(系)在Rf1a基因位点的差异,为粳型杂种优势群的利用提供基础数据。【方法】利用缺失设计引物Rfa-7F/Rfa-7R在粳稻恢复系中可扩增出957bp的片段,在保持系中可扩增出383bp的片段,对日本粳、台湾粳、韩国粳、巴西稻、美国粳、非洲IRAT粳、华北粳、非洲ITA粳、西北粳、东北粳、欧洲粳11个不同生态类型共60份材料进行PCR扩增检测。【结果】有14份材料是Rf1a/Rf1a基因类型,占所有材料的23%;日本粳、台湾粳、华北粳、西北粳和东北粳的所有材料是rf1a/rf1a基因类型,韩国粳、非洲ITA粳和欧洲粳有小部分材料是Rf1a/Rf1a基因类型,巴西粳、美国粳和非洲IRAT的大部分是Rf1a/Rf1a基因类型。【结论】粳型生态型水稻在Rf1a基因位点的差异,可以用引物Rfa-7F/Rfa-7R检测出不同基因类型,为粳型杂种优势群利用提供依据。
【Objective】Difference in Rf1 agene locus in various(series)heterotic ecotypes of japonica rice was investigated to provide basic data for utilization of heterotic groups.【Method】Deletion design primer Rfa-7F/Rfa-7Rwas adopted to amplify a segment of 957 bp in restorer and a segment of 383 bp in maintainer line and PCR amplification was carried out for 60 samples from 11 ecotypes including Japanese japonica rice,Taiwan China japonica,Korean japonica rice,Brazilian japonica rice,American japonica rice,African IRAT japonica rice,North China japonica rice,African ITA japonica rice,Northwest japonica rice,Northeast japonica rice,and European japonica rice.【Result】There were 14 samples of gene type Rf1a/Rf1 a,accounting for 23% of all samples.All samples of Japanese japonica rice,Taiwan China japonica,North China japonica rice,Northwest japonica rice,and Northeast japonica rice were gene type rf1a/rf1 a.Small part of Korean japonica rice,African ITA japonica rice and European japonica rice were gene type Rf1a/Rf1 a,while Brazil japonica rice and most of American japonica rice and African IRAT japonica rice were gene type Rf1a/Rf1 a.【Conclusion】Different gene types in ecotypes of japonica rice in Rf1 alocus can be detected by primer Rfa-7F/Rfa-7R.This provides basis for the use of heterotic ecotypes of japonica rice.
