摘要
目的研究瑶药扶芳藤乙醇、正丁醇提取物对血管内皮细胞ECV-304缺氧损伤的作用影响。方法体外培养ECV-304细胞,MTT法测定扶芳藤提取物细胞毒性,以Na2S2O4诱导ECV-304细胞缺氧损伤模型,分别将扶芳藤乙醇、正丁醇提取物作用于细胞24 h后,MTT法检测细胞存活率;收集细胞上清液,检测LDH露出率;台盼蓝计数检测细胞死亡率。结果扶芳藤乙醇和正丁醇提取物浓度为25~400μg/mL,对ECV-304细胞均无细胞毒作用;与模型对照组比较,扶芳藤乙醇(浓度50μg/mL)、正丁醇(浓度50、100、200μg/mL)提取物对缺氧损伤ECV-304细胞存活率有显著提高,差异有统计学意义(P<0.05或P<0.01)。与模型对照组比较,扶芳藤正丁醇提取物(浓度100μg/mL)能明显降低缺氧损伤ECV-304细胞上清LDH露出率,(P<0.05);与模型对照组比较,扶芳藤正丁醇提取物(浓度50、100、200μg/L)能抑制缺氧损伤细胞ECV-304凋亡(P<0.01)。结论扶芳藤提取物对Na2S2O4诱导的血管内皮细胞缺氧损伤有一定保护作用。
Objective To investigate the effect of ethanol and butanol extract of Yao medicine Euonymus fortunei on ECV-304 hypoxia injury of vascular endothelial cells. Methods ECV-304 cells were cultured in vitro, and the cytotox- icity of Euonymus fortunei extract was determined by MTT. ECV-304 cells hypoxia injury model was induced by Na2S204, respectively Euonymus fortunei ethanol and n-butanol extracts from cells after 24 h, the survival rate of cells was detected by MTT. The cell supernatant was collected for detection of LDH, the cell apoptosis rate was detected by trypan blue staining. Results Euonymus fortunei ethanol and n-butanol extract was in 25-400 txg/mL concentration range, which was no cytotoxic effect on ECV-304 cells. Compared with the model group, the ECV-304 cells survival rate of Euonymus fortunei (ethanol concentration of 50μg/mL), n-butanol (concentration of 50, 100, 200 μg/mL) extract on hypoxia injury was significantly increased (P 〈 0.05 or P 〈 0.01). Compared with the model group, the ECV-304 cells LDH exposed rate of Euonymus fortunei n-butanol extract (concentration of 100μg/mL) was significantly decreased (P 〈 0.05). Compared with the model group, the ECV-304 cells apoptosis rate of Euonymus fortunei n-butanol extract (concentration of 50, 100 and 200μg/mL) was inhibited (P 〈 0.01). Conclusion The extract of Euonymus fortunei has protective effect on Na2S204 induced hypoxia injury of vascular endothelial cells.
