摘要
目的观察凉膈散对内毒素(LPS)致急性肺损伤(ALI)大鼠肺组织病理学及血中性粒细胞(PMN)凋亡的影响。方法健康雄性SD大鼠72只,随机分为正常对照组、ALI模型组、凉膈散大、中、小剂量组和强的松治疗组,每组12只。LPS(2mg/kg)气管内滴注复制急性肺损伤模型。各组大鼠分别以生理盐水、凉膈散不同剂量、强的松灌胃。计算各组大鼠肺组织病理学积分,流式细胞仪检测PMN的凋亡改变。结果病理学炎症积分:12h模型组高于正常对照组[(2.08±0.12)分比(0.33±0.03)分,P<0.01],12h凉膈散大、中、小剂量组均低于模型组[(0.83±0.09)分、(1.00±0.09)分、(1.17±0.08)分比[(2.08±0.12)分,P<0.01或P<0.05],24h模型组高于正常对照组[(2.25±0.11)分比(0.41±0.04)分,P<0.01],凉膈散大、中、小剂量组均低于模型组[(0.67±0.05)分、(1.17±0.07)分、(1.33±0.06)分比(2.25±0.11)分,P<0.01或P<0.05]。中性粒细胞凋亡指数:12h模型组低于正常对照组[(51.36±3.99)%比(71.71±4.13)%,P<0.01],12h凉膈散大、中剂量组高于模型组[(66.37±4.25)%、(61.6±5.02)%比(51.36±3.99)%,P<0.05],24h模型组低于正常对照组[(63.04±4.89)%比(80.82±5.19)%,P<0.01],凉膈散大剂量组高于模型组[(71.32±5.32)%比(63.04±4.89)%,P<0.05]。结论凉膈散可诱导急性肺损伤时中性粒细胞凋亡,抑制中性粒细胞发挥活性作用,从而减轻急性肺损伤肺部炎症反应,其机制可能是通过抑制核转录因子-κB表达实现。
Objective To investigate the effect of Lianggesan on the histopathology accumulated points of lung tis sue and the apoptosis of polymorphonuclear granulocytes(PMN) in blood from rats with lipopolysaccharide(LPS) in duced acute lung injury. Methods Seventy-two healthy male SD rats were randomly divided into 6 groups(n=12): control group, model group, prednisone group and the Lianggesan decoction high-, middle- and low-dose groups. The LPS solution (2mg/kg) was instilled into the trachea to prepare rat acute lung injury model except control group,once a day for two days. Each treatment group completed the second infusion 1 hour after LPS solution. The drugwas intragastric administration twice a day. After each group were instilled 12h, 24h, 6 rats were put to death to collect the required samples. The apoptosis of PMN was detected with flow cytometer. Results At 12h after instilled, the pathology accumulated points in model groupwas higher than that in control group(2.08±0.12 vs 0.33±0.03, P〈0.01)), that in Lianggesan decoction high-, middle- and low-dose groups were lower than that in model group (0.83±3.09, 1.00±0.09, 1.17±0.08 vs 2.08±0.12, P〈0.01 or P〈0.05); at 24h after instilled, the points in model group was higher than that in control group(2.25±0.11 vs 0.41±0.04, P〈0.01), that in Lianggesan decoction high-, middle- and low-dose groups were lower than that in model group(0.67±0.05, 1.17±0.07, 1.33±0.06 vs 2.25±0.11, P〈0.01 or P〈0.05). The apoptosis of PMN at 12h after instilled in model group was lower than that in control group (51.36%±3.99% vs 71.71%±4.13%, P〈0.01 ), that in Lianggesan decoction high- and middle-dose groups were higher than that in model group (66.37%2 4.25%, 61.6%25.02% vs 51.36%±3.99%, P〈0.05); the apoptosis at 24h after instilled in the model groupwas lower than that in control group(63.04%±4.89% vs 80.82%±5.19%, P〈0.01), that in Lianggesan decoction high-dose groupwas higher than that in mode/ group(7
