摘要
目的:观察宣白承气汤预处理对开胸术后肺缺血再灌注损伤大鼠的肺内氧自由基的影响,从而探讨宣白承气汤的肺保护作用机制。方法:选取SPF(Specific Pathogen Free)级健康Wistar雄性大鼠30只,随机分为假手术组、模型组、治疗组,每组10只。治疗组在造模前以宣白承气汤药液灌胃3d,假手术组、模型组灌等体积的蒸馏水。假手术组开胸后仅显露左肺门但不造模,模型组及治疗组开胸后制备肺缺血再灌注损伤模型。实验结束后采集标本,测定肺湿干比值(W/D)、检测肺组织中超氧化物歧化酶(SOD)活性及丙二醛(MDA)浓度。结果:模型组大鼠肺组织W/D显著高于假手术组,治疗组显著低于模型组,差异均有统计学意义(P <0. 05);模型组SOD活性较假手术组显著降低,治疗组较模型组显著增高(P <0. 05);模型组MDA浓度较假手术组显著升高,治疗组较模型组显著降低(P <0. 05)。结论:宣白承气汤预处理可有效减少开胸手术肺缺血再灌注损伤大鼠肺内氧自由基,抑制氧化应激反应,从而起到了一定的肺保护作用。
Objective:To observe the effect of pretreatment with Xuanbai Chengqi(XBCQ)Decoction on oxyradical in lung ischemia-reperfusion injury in rat after thoracotomy,and investigate the mechanism of lung protection of XBCQ Decoction.Methods:30 healthy Wistar male rats of grade SPF were randomly divided into 3 groups:sham-operation group,model group(lung ischemia reperfusion injury group),treatment group(lung ischemia reperfusion injury+XBCQ group).Each group had 10 rats.The rats in the treatment groups with XBCQ decoction intragastrically for 3 days,the sham-operation group and model group were given equal volume distilled water.The sham-operation group only exposed the left hilum after thoracotomy but not prepare the lung ischemia reperfusion injury model.Model group and treatment group were used to prepare lung ischemia reperfusion injury model after thoracotomy.The lung wet dry ratio(W/D),superoxide dismutase(SOD)activity and malondialdehyde(MDA)concentration in lung tissue were detected at the end of the experiment.Results:The W/D in lung tissue of model group was significantly higher than that of sham-operation group,and the treatment group was lower than that of model group significantly(P<0.05).The SOD activity of model group was significantly lower than that in sham-operation group,and the treatment group was significantly higher than that in model group(P<0.05).The MDA concentration in the model group was significantly higher than that in the sham-operation group,and the treatment group was significantly lower than that in the model group(P<0.05).Conclusion:XBCQ decoction can effectively reduce oxyradical and inhibit oxidative stress in lung ischemia-reperfusion injury in rat after thoracotomy,therefore it has certain pulmonary protective effect.
