摘要
目的比较雷公藤和雷公藤加逍遥散对胶原诱导型关节炎(CIA)大鼠模型的治疗作用以及肝损伤情况。方法建立Ⅱ型胶原加完全弗氏佐剂诱导的大鼠CIA模型,将40只SD大鼠随机分为5组,空白对照组、CIA模型组、雷公藤组、低剂量逍遥散组、高剂量逍遥散组,依次记为A组、B组、C组、D组、E组,免疫加强当天,C组上午灌服生理盐水,下午灌服雷公藤药液(生药3.75 g/kg);D组上午灌服逍遥散药液(生药1.69 g/kg),下午灌服雷公藤药液(生药3.75 g/kg);E组上午灌服逍遥散药液(生药6.75 g/kg),下午灌服雷公藤药液(生药3.75 g/kg);A组与B组上午、下午均给予等体积生理盐水,给药持续28 d。以造模当日为实验第1天,实验第7天灌胃给药,每周进行1次关节肿胀度测量和AI评分,36 d处死大鼠,检测血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)的水平。结果 C组、D组、E组分别与B组比较,均可抑制大鼠的足肿胀,AI评分减少,体重增长明显(P<0.05,P<0.01);与B组比较,C组、D组、E组IL-1β、TNF-α水平显著降低(P<0.05);E组与C组、D组比较,显著抑制血清中ALT、AST水平升高(P<0.01)。结论雷公藤加高剂量逍遥散既可以治疗类风湿关节炎模型大鼠,又有对抗雷公藤致大鼠肝损伤的作用,而低剂量的逍遥散虽然不会影响雷公藤治疗类风湿关节炎的疗效,但是对雷公藤的肝毒性没有保护作用。
Objective To compare the effects between tripterygium wilfordii and the combined medication of tripterygium wilfordii and xiaoyaosan on the anti - inflammatory effect and liver injury in CIA rat models. Methods CIA rat models were prepared with Ⅱ collagen and complete Freund's adjuvant. 40 SD rats were randomized into 5 groups, a blank control group, a CIA model group, a tripterygium wilfordii group, a low- dose xiaoyaosan group and a high -dose xiaoyaosan group, labelled as A group, B group, C group, D group and E group. On the day of immune intensity, in the C group, physical saline was used in the morning, tripterygium wilfordii liquid was in the afternoon (raw herb,3.75 g/kg). In the D group, xiaoyaosan liquid (raw herb, 1.69 g/kg)was used in the morning and tripterygium wilfordii liquid was in the afternoon (raw herb,3.75 g/kg). In the E group, xiaoyaosan liquid(raw herb,6.75 g/kg)was used in the morning and tripterygium wilfordii liquid was in the afternoon( raw herb,3.75 g/kg). In the A and B groups ,the physical salien of same volume was used in the morning and the afternoon. Medication lasted for 28 days. The day of modeling was marked as the 1st day of experiment and the gavage was given on the 7th day of experiment. The joint swelling degree and AI score were measured once a week. On the 36th day,the rats were sacrificed for the determination of ALT, AST, IL - 1β and TNF - α levels in the serum. Results Compared with the B group, the paw swelling was inhibited, A1 score was reduced and body mass was increased apparently in the C, D and E groups ( P 〈 0.05, P 〈 0.01 ). Compared with the B group, the levels of IL - 115 and TNF - α were reduced significantly in the C, D and E groups (P 〈 0.05 ). Compared with the C and D groups, ALT and AST were inhibited significantly in the E group ( P 〈 0.01 ). Conclusion The combined treatment with tripterygi- um wilfordii and high - dose xiaoyaosan is effective on rheumatoid arthritis in the rat models and against liver
