UPLC同时测定鸡骨草中2种生物碱和2种黄酮碳苷的含量 被引量:6

Simultaneous determination of 2 alkaloids and 2 C-glycoside flavones in Abri Herba by UPLC

摘要 目的:建立UPLC法同时测定鸡骨草中相思子碱、下箴刺桐碱、新西兰牡荆苷2、夏佛塔苷含量,为药材质量标准改进提供方法。方法:采用HSS T3色谱柱(100 mm×2.1 mm,1.8μm),以乙腈(A)-0.2%甲酸溶液(B)为流动相进行梯度洗脱,流速0.3 m L·min^(-1),检测波长270 nm,柱温30℃。结果:相思子碱、下箴刺桐碱、新西兰牡荆苷2、夏佛塔苷的质量浓度分别在10.20~102.00μg·mL^(-1)(r=0.999 2)、6.88~68.80μg·mL^(-1)(r=0.999 0)、5.40~54.00μg·mL^(-1)(r=0.999 9)、10.92~109.20μg·mL^(-1)(r=0.999 3)的范围内线性关系良好;各成分平均加样回收率为99.1%(RSD=1.3%)、101.1%(RSD=0.69%)、103.8%(RSD=0.81%)、100.9%(RSD=0.75%)。5批样品中相思子碱、下箴刺桐碱、新西兰牡荆苷2、夏佛塔苷含量范围分别为23.02~67.24、14.98~40.09、3.02~22.51、6.34~111.38μg·g^(-1)。结论:该法经方法学验证,可同时测定鸡骨草中4种成分的含量,可为该药材的质量标准改进提供参考。 Objective: To establish a UPLC method for simultaneous determination of abrine, hypaphorine, vicenin-2 and schaftoside in Abri Herba, and to improve the quality standard of the drug. Method: Separation was performed on a HSS T3 column ( 100 mm× 2.1 mm, 1.8 μm ) and the mobile phase was aeetonitrile-0.2% formic acid with gradient elution. The flow rate was 0.3 mL · min^-1 and the wavelength was 270 nm. The column temperature was 30 ℃. Results: The linear ranges of abrine, hypaphorine, vicenin-2, schaftoside were 10.20- 102.00 μg· mL^-1 ( r=0.999 2 ), 6. 88-68.80μg· mL^-1 ( r=0.999 0 ), 5.40-54.00μg· mL^-1 ( r=0.999 9 ) and 10.92-109.20μg· mL^-1( r=0.999 3 ), respectively. The average recoveries ( n=6 ) were 99.1% ( RSD=1.3% ), 101.1% ( RSD=0.69% ), 103.8% ( RSD=0.81% ) and 100.9% ( RSD=0.75% ), respectively. The content ranges of abrine, hypaphorine, vicenin-2 and schaftoside in Abri Herba were 23.02-67.24 μg· g^-1, 14.98-40.09 μg· g^-1, 3.02-22.51 μg· g^-1 and 6.34-111.38μg· g^-1, respectively. Conclusion: Method validation suggested that the developed method was suitable for simultaneous determination of 4 components in Abri Herba, thus providing reference for the improvement of quality standard of the drug.
