摘要
目的观察姜黄素对脓毒症大鼠肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)及白细胞介素-8(IL-8)的影响。方法 30只SD大鼠,分为假手术组、脓毒症组和姜黄素治疗组,每组10只。采用改良盲肠结扎穿孔方法建立大鼠脓毒症模型,假手术组除不结扎刺穿盲肠外,其余操作同脓毒症组,姜黄素治疗组模型建立后1 h腹腔注射姜黄素(二甲亚砜作为溶剂),假手术组及脓毒症组注射同样剂量的二甲亚砜。于手术后8 h和24 h各组分别取5只大鼠用酶联免疫吸附试验(ELISA)测定血清TNF-α、IL-6、IL-8水平。结果相同时间点姜黄素治疗组的血清TNF-α、IL-6、IL-8较假手术组明显升高(P<0.05),较脓毒症组明显降低(P<0.05)。手术后24 h各组的血清TNF-α、IL-6、IL-8较手术后8 h各组的血清TNF-α、IL-6、IL-8差异无统计学意义(P>0.05)。结论姜黄素能明显抑制脓毒症时血清TNF-α、IL-6、IL-8的表达和炎症反应,早期干预对脓毒症治疗有积极意义。
Objective: To study the effects of curcumin treatment on tumor necrosis factor-α(TNF-α),interleukin-6(IL-6) and interleukin-8(IL-8) in septic rats. Methods: Thirty SD rats were randomly divided into sham operated group,septic group,and curcumin treatment group. Septic models were induced by cecal ligation and puncture(CLP). In the sham operated group,the abdomen was opened without CLP. In the curcumin treatment group,curcumin was intravenously injected into abdomina in 1 hour after CLP. Five rats of each group were sacrificed in 8 hours respectively after CLP. The others in each group were sacrificed in 24 hours after operation and their sepsis scores were recorded. The levels of TNF-α,IL-6 and IL-8 were detected by enzyme-linked immunosorbent assay(ELISA). The correlations of sepsis severity score,TNF-α,IL-6 and IL-8 were analyzed. Results: The levels of TNF-α,IL-6 and IL-8 of the curcumin treatment group were significantly higher than the control group(P〈0.05),while significantly lower than septic group(P〈0.05). The difference on the levels of TNF-α,IL-6 and IL-8 among the groups in 24 hours after operation showed no significant difference with the results in 8 hours after operation(P〈0.05). Conclusion: Curcumin can significantly inhibit the expression of serum TNF-α,IL-6,IL-8,and inflammatory reaction in sepsis. The early intervention has a positive effect on the treatment of sepsis.
