摘要
CpG ODN是人工合成非甲基化的CpG寡聚脱氧核苷酸链,可以与B细胞、树突状细胞、单核细胞和巨噬细胞等免疫细胞中内质网膜上的Toll样受体9(TLR9)结合,是TLR9受体激动剂。CpG ODN是一种具有促进Th1免疫应答特点的疫苗佐剂,能够提高疫苗的免疫原性并且具有较好的安全性。对感染、肿瘤和过敏等疾病的预防和治疗具有应用前景。本文主要从CpG ODN简介、分类及结构特性、免疫机制、安全性和疫苗佐剂在临床中应用方面展开,为进一步的研究和应用CpG ODN佐剂提供参考。
CpG ODN, a synthetic oligodeoxynucleotide containing CpG motifs, is a TLR9 agonist as it could be recognized by Toll-like receptor 9 (TLR9) expressed on the membranes of B cells, dendritic cells, monocytes and macrophages. CpG ODN is the kind of vaccine adjuvant that promotes Thl immune re- sponses and enhances the immunogenicity of vaccines with high safety, showing promising prospects in the prevention and treatment of infections, tumors and allergic diseases. This review mainly focuses on the intro- duction, classification and structure features, immune mechanism, safety and clinical application of CpG ODN as vaccine adjuvant in order to provide references for further researches and application of CpG ODN as immune adjuvant.
