摘要
目的分析晚期糖基化终末产物(AGEs)的表达与2型糖尿病大鼠肺纤维化的相关性。方法随机选取15只大鼠予高糖高脂饮食及腹腔注射链脲佐菌素诱导建立2型糖尿病模型(糖尿病组),余15只作为正常对照组。采用苏木素一伊红(HE)、Masson染色法对大鼠肺组织进行染色,观察模型建立12周后肺脏形态学的改变及胶原纤维的分布情况;应用免疫组化检测肺组织中AGEs的表达部位及水平。结果糖尿病组大鼠肺间质中成纤维细胞显著增多,并可见胶原纤维和弹性纤维沉积,AGEs在糖尿病组大鼠肺组织细胞中表达均明显增强,差异均有统计学意义(P〈0.01)。结论2型糖尿病大鼠肺纤维化程度与肺组织AGEs表达增强具有相关性,提示AGEs可能参与了糖尿病肺纤维化的发生发展。
Objective To analyze the correlation between the expression of advanced glycation end products (AGEs) and pulmonary fibrosis of type 2 diabetic rats. Methods Fifteen rats were randomly selected as the diabetes group and the remaining 15 rats belonged to normal control group. The model of type 2 diabetic rats was induced by injecting strep-tozotocin along with high sugar and high fat diet. The changes of morphology and the collagen accumulation in the lung of model rats treated by HE and Masson staining were observed after 12 weeks, and the distribution and protein level of AGEs expressed in lung tissues were detected by immunohistochemistry. Results As the fibroblasts increased and collagen fiber hyperplasia in interstitial lung in diabetes group,the expression of AGEs also significantly increased with statistical significance(P 〈 0.01 ). Conclusion The pulmonary fibrosis degree of diabetic rats is related to the enhanced expression of AGEs in lung tissue, which suggests that AGEs might be involved in the occurrence and development of diabetic pulmonary fibrosis.
