摘要
为研究番茄红素对山羊精液冷冻保存效果的影响,在山羊精液冷冻稀释液中添加不同质量浓度(0、0.5、1.0、2.0、4.0mg/mL)的番茄红素,检测冷冻-解冻后精子的活率、顶体完整率、质膜完整率、线粒体活性,并用试剂盒测定超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)的活性。结果表明,1.0mg/mL组的冷冻保存效果最好,精子活率、顶体完整率、质膜完整率和线粒体活性分别达到52.04%、51.33%、53.57%和49.31%,显著高于对照组,且SOD、CAT、GSH-Px的活性也显著高于对照组;0.5mg/mL组与2.0mg/mL组的精子活率、顶体完整率、质膜完整率、线粒体活性及CAT和GSH-Px活性差异不显著;4.0mg/mL组的精子活率、顶体完整率、质膜完整率、线粒体活性及SOD和CAT活性与对照组差异不显著。因此,在山羊精液冷冻稀释液中加入1.0mg/mL的番茄红素对山羊精液具有显著的保护作用,能进一步提高山羊冻精品质。
The aim of this study is to determine the effects of different amounts of lycopene (0 mg/mL,0.5 mg/mL,1.0 mg/mL,2.0 mg/mL,4.0 mg/mL) added to cryopreservation extender on the post-thawed sperm parameters (motilities,acrosome integrity,plasma integrity and mitoehondrial activity) and antioxidant activities (SOD,CAT and GSH-Px) of sperm of white cashmere goat from Shanbei.The results showed that addition of lycopene was 1.0 mg/mL,the motility (52.04%),acrosome integrity (51.33%),plasma membrane integrity (53.57%) and mitochondrial activity (49.31%) were increased significantly and also had significant differences in enzyme activity of SOD,GSH-Px and CAT compared with the control group.For 0.5 mg/mL group and 2.0 mg/mL group,the motility,acrosome integrity,plasma membrane integrity,mitochondrial activity and CAT,GSH-Px activity were no significant difference between them.For 4.0 mg/mL group,the motility,acrosome integrity,plasma membrane integrity,mitochondrial activity and SOD,CAT activity were significantly lower than that of control group.The results of this study showed that lycopene supplementation in semen extenders was of greater benefit to sperm of frozen-thawed goat sperm,the best mass concentration was 1.0 mg/mL.
