摘要
目的研究蜘蛛香Valeriana jatamansi Jones的化学成分及其神经保护活性。方法采用硅胶、Sephadex LH-20、ODS、HPLC制备柱,对蜘蛛香75%乙醇提取物的石油醚部位进行分离纯化,通过波谱数据鉴定所得化合物的结构。再观察其对CoCl_2诱导大鼠嗜铬细胞瘤(PC12)细胞凋亡的抑制作用。结果从中分得11个化合物,分别鉴定为蜂斗菜内酯B(1)、蜂斗菜内酯D(2)、jatamanvaltrate E(3)、jatamanvaltrate A(4)、valtrate hydrine B8(5)、valeriotetrate C(6)、valtral C(7)、chlorovaltrate A(8)、β-谷甾醇(9)、β-谷甾醇乙酸酯(10)、棕榈酸(11)。化合物2、4、6、8能显著抑制PC12细胞凋亡(P<0.05)。结论化合物2为首次从败酱科植物中分离得到,化合物10为首次从该植物中分离得到,化合物2、4、6、8具有神经保护活性。
AIM To investigate the chemical constituents from Valeriana jatamansi Jones and their neuroprotective activities. METHODS The petroleum ether fraction of 75% ethanol extract from V. jatamansi was isolated and purified by silica, Sephadex LH-20, ODS and preparative HPLC column, the structures of obtained compounds were elucidated by spectral data. Then their inhibitory effects on the apoptosis of rat pheochromocytoma (PC12) cells induced by CoC12were observed. RESULTS Eleven compounds were isolated and identified as bakkenolide B (1), bakkenolide D (2), jatamanvahrate E (3), jatamanvaltrate A (4), valtrate hydrine B8 (5), valeriotetrate C (6), valtral C (7), chlorovahrate A (8), β-sitosterol (9), β-sitosterol acetate ( 10 ), palmitic acid ( 11 ). Compounds 2, 4, 6 and 8 could significantly inhibit the apoptosis of PC12 cells ( P 〈 0.05 ). CONCLUSION Compound 2 is isolated from genus Valerianaceae for the first time, compound 10 is first obtained from this plant, and compounds 2, 4, 6 and 8 have neuroprotective activities.
