摘要
目的:观察表没食子儿茶素没食子酸酯(epigallocatechin-3-gallate,EGCG)对甲状腺癌细胞增殖和摄碘能力的影响。方法:用不同浓度的EGCG处理甲状腺癌W3细胞株48 h后,用3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐(MTT)法检测W3细胞增殖能力,流式细胞术、免疫荧光法检测钠-碘转运体(sodium iodide symporter,NIS)的变化,摄碘能力实验检测W3细胞摄碘能力。结果:与对照组比较,随着EGCG浓度的增加,W3细胞的增殖能力逐渐降低(P<0.01),NIS的表达逐渐增加(P<0.01),呈浓度依赖性。摄碘实验显示,在一定浓度范围内,EGCG明显促进W3细胞对碘的吸收能力(P<0.01)。结论:EGCG能抑制甲状腺癌细胞的增殖能力,通过上调NIS的表达促进甲状腺癌细胞株的摄碘能力。
Objective: To explore the effect of epigallocatechin-3-gallate (EGCG) on the proliferation and radioiodine accumulation in papillary thyroid cancer(PTC) cells. Methods: After PTC cells W3 were treated by using different concentrations of EGCG for 48 h, the proliferation of PTC cells was detected by 3- 4,5-dimethylthiazol-2-yl-2,5-diphenyl-tetrazolium bromide (MTF) assay ; the expression of sodium iodide symporter(NIS) in PTC cells was detected by flowcytometry and immunofluorescence methods; the ability of PTC cells to iodide-uptake was measured by iodide-uptake assay. Results: The proliferation of PTC cells was decreased, and the expression of NIS in VI'C cells was increased, in a concentration dependent manner. And the ability of iodide-uptake in PTC cells was increased significantly in some concentration gradient. Conclusion: EGCG inhibited the growth of PTC cells and enhanced the ability of iodide-uptake through increased expression of NIS.
