摘要
目的:探讨赤芍不同给药剂量对大鼠脂多糖(Lipopolysaccharide,LPS)诱导的急性肺损伤(acute lung injury,ALI)的预防作用,筛选最佳给药剂量,为后期药效物质基础研究奠定基础。方法:SD大鼠随机分为高剂量组、中剂量组、低剂量组、阳性药(DEX)组、模型组(LPS组)、空白组(NS组),每组10只。采用气管内滴注内毒素方法^([1])建立急性肺损伤模型,造模12h后取肺灌洗液检测白细胞数目、蛋白浓度,取右肺上叶测湿干比,取右肺下叶观察肺组织病理形态学变化。结果:地塞米松、赤芍高中剂量给药组均能显著减少白细胞数目(P<0.05),降低肺湿干比(W/D)(P<0.05)和蛋白浓度(P<0.05),缓解肺水肿、肺泡结构破坏、炎性细胞浸润,改善肺内出血情况。结论:赤芍能显著改善肺部结构受损、肺水肿、中性粒细胞浸润等病变,尤其以中剂量为佳。
Objective: To probe the preventive effect of different doses of Paeoniae Rubra Radix on lipopolysaccharide-induced acute lung injury in rats,filtering the optimal dose and speculating the possible mechanism.Methods: SD rats were divided into a high dose group,a middle dose group,a low dose group,a positive drug group( DEX group),a model group( LPS group) and a control group( NS group),with 10 rats in each group. The model group was established by intratracheal administration of lipopolysaccharide. 12 h later detect the number of white blood cells,protein concentration from the lung lavage fluid and measure wet weight and dry weight of upper lobe of the right lung and observe pathological changes of lower lobe of the right lung. Results: Dexamethasone,the high dose group and the middle dose group could significantly reduce the number of white blood cells( P〈0. 05),reduce lung wet to dry ratio( W / D)( P〈0. 05) and protein concentration( P〈0. 05),relieve pulmonary edema,alveolar structural damage and inflammatory cell infiltration and reduce pulmonary hemorrhage. Conclusion: Radix paeoniae rubra can relieve the acute lung injury induced by lipopolysaccharide and the middle dose is the optimal.
