摘要
目的:通过观察不同浓度通关藤提取物对人胃腺癌BGC-823细胞增殖、凋亡及MTDH基因表达的影响,为胃癌的临床治疗提供实验基础及潜在治疗靶点。方法:(1)MTT法检测通关藤提取物对BGC-823细胞增殖影响。(2)Annexin V/PI双染法检测通关藤提取物对BGC-823细胞凋亡影响。(3)反转录多聚酶链反应(RTPCR)检测通关藤提取物对BGC-823细胞MTDH mRNA表达影响。(4)Western Blot法检测通关藤提取物对BGC-823细胞凋亡相关蛋白表达影响。结果:(1)通关藤提取物作用于BGC-823细胞,细胞生长抑制率随药物浓度增大而升高,加药各组与对照组相比,差异有统计学意义。(2)通关藤提取物0 mmol/L、0.5 mmol/L、1 mmol/L、2mmol/L诱导BGC-823细胞凋亡率为(2.98±1.01)%、(6.79±0.68)、(12.512±1.27)%、(16.97±1.09),与对照组相比差异有统计学意义。(3)通关藤提取物可下调BGC-823细胞MTDH mRNA表达,这种作用具有浓度依赖性。(4)通关藤提取物能够下调Bcl-2家族中的抑凋亡因子Bcl-2、Bcl-xl的表达尤其是Bcl-xl的表达,上调凋亡执行者Caspase-3的表达。结论:通关藤提取物可下调BGC-823细胞MTDH mRNA表达,并通过下调Bcl-2、Bcl-xl,上调Caspase-3的表达而诱导BGC-823细胞凋亡,从而抑制了BGC-823细胞的生长增殖。
Objective: To investigate the effects of different concentrations of Marsdenia tenacissima extract on human gastric cancer BGC-823 cells proliferation,apoptosis and effect on the expression of MTDH gene,for the clinical treatment of gastric cancer provide an important experimental basis and potential therapeutic targets. Methods:①MTT-assay was used to detect the effect of the extract of the extract on the proliferation of BGC-823 cells.②The expression of BGC-823 cells apoptosis were studied using Annexin V/PI.③The expression of the expression of mRNA BGC-823 in MTDH cells were studied using flow cytometry and fluorescence realtime quantitative RT-PCR. Results:① Extract from Marsdenia Tenacissima acting on the BGC-823 cells,with the increasing concentration of drug increased growth inhibition rate in each group compared with the control group,the difference was statistically significant.②Extract from Marsdenia Tenacissima concentration 0 mmol/L,0. 5 mmol/L,1 mmol/L,2 mmol/L,BGC-823 induced apoptosis rate was( 2. 98 ± 1. 01) %,( 6. 79 ± 0. 68),( 12. 512 ± 1. 27) %,( 16. 97 ± 1. 09),compared with the control group,the difference was statistically significant.③Extract from Marsdenia Tenacissima down-regulated mRNA expression in BGC-823 cells MTDH,this effect is dose-dependent.④Extract from Marsdenia Tenacissima downregulates Bcl-2 expression of Bcl-xl expression,especially anti-apoptotic factor,Bcl-xl in the Bcl-2 family,were up-regulated the ex-pression of apoptosis execution of Caspase-3. Conclusion: Extract from Marsdenia Tenacissima reduced mRNA expression in BGC-823 cells MTDH,and by down-regulating Bcl-2,Bcl-xl,regulate the expression of Caspase-3,induce apoptosis in BGC-823 cells,thereby inhibiting the BGC-823 cells proliferation.
