摘要
研究对安徽省四个地方鸡种遗传资源淮南麻黄鸡、霍邱鸡(固始鸡)、黄山黑鸡、金寨黑鸡的遗传多样性和群体遗传结构进行了分析。利用基因组扫描技术检测21个微卫星标记在四个群体中的基因型,统计分析各群体的遗传多样性指标。结果显示:21个微卫星标记在四个群体中表现出中高度多态性;观察杂合度最高的是金寨黑鸡,最低的是霍邱鸡,分别为0.621和0.587;Nei′s标准遗传距离最近的是淮南麻黄鸡和霍邱鸡,为0.0212,最远的是淮南麻黄鸡与黄山黑鸡,为0.2187;聚类分析显示淮南麻黄鸡和霍邱鸡聚为一类,黄山黑鸡和金寨黑鸡聚为一类;遗传结构分析表明淮南麻黄鸡和霍邱鸡群体极为相似,黄山黑鸡和金寨黑鸡有较大差异。安徽四个地方鸡种均表现出较高的遗传多样性,黄山黑鸡与金寨黑鸡遗传结构有较大差异,淮南麻黄鸡与霍邱鸡非常相似,在以后的选育开发中应充分利用这些遗传特性。
This study was aimed to estimate the genetic diversity and population structure of four kinds of Anhui local chicken breeds by 21 microsatellite markers. A total of 370 individuals originating from 4 Anhui local chicken populations(Huainan partridge chicken,HN;Huoqiu chicken,HQ;Huangshan black chicken,HS;Jinzhai black chicken,JZ)were analyzed in this study. The results showed that the lowest estimate of observed heterozygosity(0.587)was obtained in HQ,while the highest one(0.621)was found in JZ. The Nei′s genetic distance values ranged from 0.0212(between HN and HQ)to 0.2187(between HN and HS). A phylogenetic consensus tree indicated that the 4 Anhui local chicken populations were divided into two categories. The results of the clustering analysis indicated that the population structures of HN and HQ were similar,the population structures of HS and JZ were different. The results demonstrated that high genetic diversity was observed in the 4 Anhui local chicken populations,and it would play an important role in conservation,supervision,and utilization of Anhui local chicken resources.
