摘要
目的选择均质法从侧柏叶中提取黄酮类化合物。方法采用比色法,以芦丁为对照品,建立了含量测定法。采用单因素试验法,对比超声、微波、回流和均质提取法,并利用正交试验,以均质时间、甲醇浓度、料液比和次数4个因素进行考察,结果总黄酮浓度在0.008 96~0.053 76 mg/m L范围内呈良好的线性关系,加样回收率为102.07,RSD为2.77%。正交试验结果:均质时间15 min,甲醇浓度为70%,料液比1∶10,提取1次。结论所建立的含量测定法简单易行,均质法比其他方法更为简便,省去对物料粉碎的步骤,且正交试验确定的试验结果稳定、可靠,可用于工业生产。
Objective To extract the flavonoids from Cacumen platycladi by homogeneous method.Methods The content determination method was established by colorimetric method with rutin as the reference substance.The ultrasound,microwave,reflux and homogeneous extraction method were compared using the single factor experiment method,and the orthogonal test was used with homogeneous time,methanol concentration,solid4 iquid ratio and times as four factors were.Results Total flavonoids had good linear relationship in the range of 0.008 96~0.053 76 mg/mL,the average recovery rate was 102.07 and RSD was 2.77%.Orthogonal test results were as follows:the homogeneous time was 15 min,methanol concentration was 70%,and the material-liquid ratio was 1:10,extraction for 1 time.Conclusion The established method for content determination is simple and feasible,in which the homogeneous method is simpler,and it can save the procedure of material crush.The established orthogonal test has stable and reliable results,which can be used in industry production.
