摘要
目的探讨新型钌卟啉化合物(Rup)-03、Rup-04对高分化鼻咽癌细胞(CNE)-1、低分化CNE-2和结肠癌SW480细胞体外增殖和凋亡的影响。方法经光照和不同浓度(10-10mol/L、10-9mol/L、10-8mol/L、10-7mol/L、10-6mol/L、10-5mol/L)Rup-03或Rup-04联合作用后,联合磺酞罗丹明B法和吉姆萨染色法观察CNE-1、CNE-2和SW480细胞的形态学变化及其增殖和凋亡的情况,邻苯三酚自氧化法和Griess法测定其超氧阴离子和NO表达。结果随着Rup-03、Rup-04作用浓度的升高,CNE-1、CNE-2和SW480细胞凋亡增多,对超氧阴离子的清除能力降低,细胞内NO表达升高(均P<0.05),且Rup-03对3种肿瘤细胞的光动力抑制作用均强于Rup-04(P<0.05)。结论 Rup-03和Rup-04均对CNE-1、CNE-2和SW480肿瘤细胞有浓度依赖性的促凋亡作用,其抗肿瘤机制可能与Rup-03、Rup-04产生自由基诱导细胞凋亡有关。
Objective To explore the effect of novel ruthenium porphyrin complex ( Rup)-03 and Rup-04 on in vitro proliferation and apoptosis of well-differentiated nasopharyngeal carcinoma cell ( CNE)-1,poorly-differentiated CNE-2,and colon cancer cell SW480. Methods After the intervention of photodynamic therapy and Rup-03/Rup-04 with different concentrations ( 10 -10 mol/L,10 -9 mol/L, 10 -8 mol/L,10 -7 mol/L,10 -6 mol/L and 10 -5 mol/L),correspondingly,the morphologic changes,proliferation and apoptosis of CNE-1, CNE-2,and SW480 were observed using sulforhodamine B assay and Giemsa staining ,the expressions of superoxide anion and NO of the cells were determined using pyrogallol autoxidation method and Griess assay .Results For CNE -1,CNE-2,and SW480,the cell apoptosis increased ,superoxide anion scavenging activity decreased ,and the intracellular NO expressions increased with the increase of the intervention concentrations of Rup-03/Rup0-4 (all P〈0.05).The inhibitory effect of Rup0-3 on photodynamics of three tumor cells were stronger compared to Rup-04(P〈0.05).Conclusion Both Rup-03 and Rup-04 have concentration-dependent pro-apoptotic effect on tumor cells including CNE-1,CNE-2 and SW480.The antit-umor mechanism may be related to the apoptosis induced by Rup -03/Rup-04-produced radicals.
