摘要
对参连方中醇提部分工艺进行优选。在以致豚鼠心律失常时哇巴因的消耗量为药效指标,考察全方水提,全方水提醇沉,党参等水提醇沉,丹参等醇提和党参等水提、丹参等醇提4种不同提取工艺路线的基础上,采用L9(34)正交表进行正交实验,以不同实验方案的出膏率及丹酚酸B、丹参酮Ⅱ_A、小檗碱3种指标成分的含量为评价指标,研究乙醇浓度、乙醇用量、提取时间、提取次数4个因素对醇提部分提取工艺的影响,应用综合评分法优选最佳提取工艺,并进行工艺验证试验。所得最佳提取工艺为加入60%乙醇共提取2次,分别加入10,9倍量、提取2.0 h,丹酚酸B、丹参酮Ⅱ_A、小檗碱提取量分别为42.01,0.41,40.15 mg·g^(-1)。优选的工艺稳定、可行,可为参连颗粒的工业化生产提供依据。
To optimize the ethanol extraction process for Shenlian formula. On the basis of the pharmacodynamics index for different extraction process routes,the contents of salvianolic acid B,tanshinone ⅡAand berberine,as well as the extraction ratio in different experimental schemes were used as the ethanol extraction examining indexes,and multi-criterion synthesizing grading method was used for data analysis to optimize and verify the ethanol-extraction process conditions in orthogonal experiment. The optimum ethanol extraction process was as follows: adding 60% ethanol,10 times amount,extracting for 2. 0 h each time for a total of 2 times. This extraction process showed good stability and availability.
