摘要
目的:探讨枸杞多糖(LBP)联合替莫唑胺(TMZ)治疗对大鼠脑胶质瘤的作用机制。方法:将SD大鼠分为空白对照组、模型组、TMZ组和LBP+TMZ组,除空白对照组外,其他3组分别于颅内接种C6脑胶质瘤细胞。TMZ组给予TMZ灌胃,LBP+TMZ组给予TMZ及LBP分别灌胃,空白对照组和模型组给予等量生理盐水。各组干预28d后测量肿瘤体积,采用酶联免疫吸附试验(ELISA)法检测血清白介素-17(IL-17)水平,RT-PCR检测Foxp3mRNA相对表达量,流式细胞术检测C6脑胶质瘤组织Th17、Treg细胞比率。体外培养C6脑胶质瘤细胞,分为模型组、TMZ组和LBP+TMZ组,培养72h后检测细胞上清液IL-17水平和细胞Foxp3mRNA相对表达量。结果:治疗后,LBP+TMZ组的肿瘤体积显著小于TMZ组(P<0.05)。LBP+TMZ组IL-17水平显著低于TMZ组(P<0.05)。LBP+TMZ组Foxp3mRNA相对表达量较模型组、TMZ组显著降低(P<0.05)。LBP+TMZ组Treg细胞比率较TMZ组减少(P<0.05),Th17/Treg比值显著高于模型组和TMZ组(P<0.05)。结论:LBP联合TMZ可有效抑制大鼠脑胶质瘤,其机制可能与调节免疫细胞Th17、Treg的表达有关。
Objective: To explore the effects and mechanisms of combined lyceum hararum polysaccharides(LBP) with temozolomide (TMZ) on brain glioma in rats. Methods: SD rats were divided into 4 groups: control group, model group, TMZ groupand LBP+TMZ group. The rats were intracranially inoculated with C6 glioma ceils except for the con- trois. Rats in the treatment groups were administered TMZ or LBP plus TMZ via intragastric gavage, while those in the control and model groups were administered equal volume of normal saline. After 28 days administration, the rats were sacrificed and the tumor size was tested. The serum IL-17 level and Foxp3 mRNA expression were determined by enzyme-linked immuno sorbent assay (ELISA) and RT- PCR, respectively. The percentages of Th17 and Treg cells in glioma tissue were examined by flow cytom- etry. C6 glioma cells were subsequently cultured in vitro and divided into 3 groups:model group, TMZ group and LBP+TMZ group. After 72 h of cultivation, the IL-17 level in ceil supernate and Foxp3 mRNA expression were also detected. Results: The size of tumors in the LBP+TMZ group were notably smaller than those in the TMZ group ( P〈0.05). The IL-17 level in rat serum and C6 glioma cells supernate were lower in the LBP+TMZ group than those in the TMZ group ( P〈0.05). The Foxp3 mRNA expression was down-regulated in the LBP + TMZ group than those in the model group and the TMZ group ( P〈0. 05). The percentage of Treg cells was obviously reduced in the LBP+TMZ group compared to the TMZ group ( P〈0.05), and the ratio of Th17/Treg was significantly lower than those in the model group and the TMZ group ( P〈0. 05). Conclusion: A combination administration of LBP and TMZ could effectively suppress the growth of brain glioma in rats via modulating Th17 and Treg cells.
