摘要
目的研究绿原酸和甘草酸联合应用体外抗呼吸道合胞病毒作用。方法以利巴韦林为阳性药,采用细胞病变抑制试验,噻唑蓝比色法检测细胞活性,观察绿原酸和甘草酸单用与合用不同比例(3∶1、2∶1、1∶1、1∶2)在Hep-2细胞中对呼吸道合胞病毒的抑制作用。结果在不同方式作用下,与绿原酸、甘草酸单独用药相比,联合组治疗指数增大,最大无毒浓度时病毒滴度小于单独用药组,抑制呼吸道合胞病毒的作用显著增强。当配比为绿原酸(133μg·ml^(-1))+甘草酸(67μg·ml^(-1))2∶1时,对呼吸道合胞病毒抑制率最大,治疗指数最高,病毒滴度最小。结论绿原酸与甘草酸联合用药对呼吸道合胞病毒的抑制具有协同增效作用,增强了体外抗呼吸道合胞病毒作用。
Objective To investigate the anti-respiratory syncytial virus effects of chlorogenic acid combined with glycyrrhizin in vitro . Methods The inhibitory effect of chlorogenic acid combined with glycyrrhizin at different ratios(3 : 1,2 : 1,1 :1,1 : 2) on respiratory syncytial virus(RSV) in Hep-2 cells was observed using eytopathic inhibition assay and MTT assay with ribavirin as the positive drug. Results The combined therapeutic index (TI) of ehlorogenic acid and glyeyrrhizin increased, the virus titers decreased at the maximum non-toxic concentration, and the inhibitory effect on RSV significantly enhanced compared with the monotherapy of chlorogenic acid and glycyrrhizin. In addition, when the ratio of chlorogenic acid (133μg·ml^-1) to glyeyrrhizin (67μg·ml^-1) was 2 :1, the inhibitory rate of ehlorogenic acid and glycyrrhizin on RSV was maximal, TI was the highest and the virus titers were minimal. Conclusion Combined application of chlorogenic acid and glycyrrhizin has a synergistic inhibitory effect on RSV and can enhance the anti respiratory syncytial virus effect in vitro .
