摘要
目的:探讨他扎罗汀(Tazarotene)对角质形成细胞增殖周期蛋白Cyclin D1、CDK4和CDK6表达水平的影响。方法:分别以不同浓度的他扎罗汀作用于HaCaT细胞,用CCK-8比色法检测每组细胞在12、24、48和72 h的增殖情况,流式细胞仪检测细胞周期变化,western blot检测周期相关蛋白水平变化。结果:他扎罗汀对HaCaT细胞具有明显的增殖抑制作用(F=21.23,P<0.05),并使G0/G1期细胞比例增加(F=19.83,P<0.05),周期相关蛋白Cyclin D1、CDK4、CDK6的表达水平明显下调。结论:他扎罗汀对HaCaT细胞具有明显的增殖抑制和周期阻滞作用,其作用机制可能与下调Cyclin D1/CDK4/CDK6蛋白复合物的表达有关。
Objective: To investigate the effects of tazarotene on the expression of Cyclin D1/CDK4/CDK6 in keratinocytes. Methods: Keratinocytes(HaCaT) were treated with different concentrations of tazarotene. Cell proliferation was determined with CCK-8 assay after 12, 24, 48 and 72h of treatment. Changes in cell cycle and cycle related protein levels were assessed with flow cytometry and western blot, respectively. Results: Tazarotene dramatically inhibited proliferation of HaCaT cells in a dose-de- pendent manner(F=21.23, P〈0.05), and increased the proportion of cells in G0/G1 phase(F=19.83, P〈0.05). Tazarotene signifi- cantly decreased expression levels of Cyclin D1, CDK4 and CDK6 proteins. Conclusion: Tazarotene inhibits keratinocyte prolif- eration and cell cycle progression, possibly via down regulation of expression levels of Cyclin D1/CDK4/CDK6 protein complex.
