摘要
目的建立超高效液相色谱-荧光法(UPLC-FLD)测定婴幼儿乳粉中胆碱含量的方法。方法利用1-萘基异氰酸酯与胆碱反应生成稳定的、具有荧光的阳离子芳香族化合物,用超高效液相色谱-荧光法分离测定。乳粉样品经盐酸水解,无水硫酸钠脱水后与1-萘基异氰酸酯进行衍生反应,经ACQUITY UPLC BEH HILIC色谱柱分离,用流动相四甲基氢氧化铵-羟基乙酸-乙腈-水溶液进行洗脱,然后通过荧光检测器(FLD)进行检测,激发波长和发射波长分别为220 nm和350 nm。结果胆碱的浓度(以胆碱阳离子计)在0.90μg/ml^13.5μg/ml时线性关系良好(相关系数r=0.999 7),回收率为89.0%~92.5%,仪器的检出限为0.052μg/ml。结论该方法具有成本低、灵敏度高、快速、简便等优点。
Objective To develop a method for the determination of choline in infant milk powder by ultra- high performance liquid chromatography with ultra- high performance liquid chromatography with fluorescence detection( UPLC- FLD). Methods Infant milk powder was firstly hydrolyzed with hydrochloric acid,then dehydrated with anhydrous sodium sulfate and derivatized with 1- naphthyl isocyanate,and finally the choline derivative was seperated by UPLC using ACQUITY UPLC BEH HILIC column with a mobile phase contained tetramethylammonium glycolate buffer,and determined by fluorescence detection with the excitation wavelenghth of 220 nm and emission wavelength of 350 nm. Results Good linearity was obtained when the concentration of choline was within 0. 90 μg / ml- 13. 5 μg / ml. The relative coefficient( r) was 0. 999 7. The recoveries were within 89. 0%- 92. 5%. The detection limit was 0. 052 μg / ml. Conclusion The method is simple,sensitive,quick and inexpensive.
