电针“大椎”“命门”对脊髓损伤大鼠神经元细胞凋亡及JNK信号通路相关蛋白表达的影响 被引量:48

Effect of Electroacupuncture Stimulation at“Dazhui”(GV 14)and“Mingmen”(GV 4)on Cell Apoptosis of Spinal Cord and Expression of JNK Signaling Related Protein in Spinal Cord Injury Rats

摘要 目的:观察督脉电针对脊髓损伤(SCI)大鼠不同时间窗下神经元细胞凋亡及磷酸化c-Jun氨基末端激酶(p-c-Jun)表达的影响,探讨督脉电针在脊髓损伤与修复过程中的作用机制。方法:SD大鼠随机分为3个时间窗组(术后1、3、7d组),每个时间窗组再随机分3个组:正常对照组、模型组和督脉电针组。采用改良式的Allens打击法复制脊髓损伤模型。督脉电针组选取"大椎""命门"进行电针干预,每日1次,每次20min,连续治疗至相应天数。采用BBB评分法评估大鼠后肢运动神经功能的变化,TUNEL法检测各组大鼠受损脊髓神经元细胞凋亡数量,Western blot法检测各组脊髓损伤组织中p-c-Jun蛋白的表达情况。结果:BBB评分结果显示,模型组评分显著低于正常对照组(P<0.01),术后3、7d督脉电针组BBB分值均高于模型组(P<0.05,P<0.01)。与正常对照组相比,在3个时间窗下,模型组大鼠受损脊髓组织内凋亡阳性细胞数目均显著升高(P<0.01);与模型组相比,在3个时间窗下,督脉电针组较模型组凋亡细胞显著减少(P<0.01)。与正常对照组相比,在3个时间窗下,模型组大鼠受损脊髓组织内p-c-Jun蛋白表达均显著升高(P<0.05,P<0.01);与模型组相比,术后3、7d督脉电针组大鼠受损脊髓组织中p-c-Jun表达显著降低(P<0.01,P<0.05)。结论:脊髓损伤后受损脊髓组织中p-c-Jun蛋白表达水平升高;督脉电针可以改善脊髓损伤大鼠运动神经功能,其对脊髓损伤的保护作用机制可能与下调受损脊髓组织中p-c-Jun蛋白表达有关。 Objective To observe the effect of electroacupuncture(EA)stimulation at"Dazhui"(GV 14)and"Mingmen"(GV 4)of the Governor Vessel at different time-points on spinal cord neuronal apoptosis and the expression of c-Jun N-terminal kinases(JNK)protein in spinal cord injury(SCI)rats,so as to reveal its mechanism underlying improving SCI.Methods A total of 108 male SD rats were randomly divided into normal control,SCI model and EA groups which were further divided into 1,3and 7dsubgroups(12rats/subgroup,6rats in each subgroup for TUNEL or Western blot,separately).SCI model was established by using the modified Allen's method.EA was applied to GV 14 and GV 4for 20 min,once daily,for 1,3and 7days,respectively.Basso-Beattie-Bresnahan(BBB)scale was adopted to assess the locomotor function of rats,the TUNEL method was used to examine neuronal apoptosis of injuried spinal cord,and the expression of phosphorylated(p)-c-Jun protein of T9-T11 spinal cord was detected by using Western blot.Results After modeling,the BBB scores of SCI rats on day 1,3and 7were significantly decreased(P0.01),while the numbers of apoptotic neuronal cells and the expression levels of p-c-Jun protein in the spinal cord were considerably increased at the 3time-points in the model group(P0.01,P0.05).Following EA intervention,the decreased BBB scores on day 3and 7,and the increased numbers of apoptotic neuronal cells on day 1,3and 7and the upregulated expression levels of p-c-Jun protein on day 3and 7were obviously suppressed(P0.05,P0.01).ConclusionEA intervention can improve the locomotor function of SCI rats,which may be related to its effects in reducing neuronal apoptosis and down-regulating p-c-Jun protein in the injuried spinal cord.
