眼眶静脉采血及尾静脉注射给药对大鼠肝细胞内NF-κB的影响 被引量:1

摘要 目的研究常规动物实验操作对大鼠肝细胞内核转录因子Kappa B(简称NF-κB)的影响。方法采用雄性SD大鼠,分别进行眼眶静脉采血、尾静脉注射操作,每天记录大鼠体质量和饲料消耗数据,连续处理14 d后,测定各组大鼠肝细胞内NF-κB p65的mRNA表达量和蛋白水平,并与对照组进行比较,分析这些操作的影响。结果与对照组相比,眼眶静脉采血及尾静脉注射实验组大鼠体增重和饲料能耗比均有降低,且差异极显著(P<0.01);实验组大鼠肝细胞中NF-κB p65和蛋白的表达水平较对照组都有升高,且差异极显著(P<0.01);实验组中眼眶静脉采血组NF-κB p65和蛋白的表达水平高于尾静脉注射组,差异极显著(P<0.01)。结论本研究中涉及的常规实验操作均引起大鼠的显著应激反应致福利受损,可导致肝细胞内NF-κB表达水平提高,表明了NF-κB参与了大鼠福利损伤的过程。 Objective To study the expression of NF-κB in rats' liver after welfare impaired by experimental procedures. Method Male SD rats, welfare were impaired by experimental procedures. Body weight gain and food consumption were recorded throughout the study , mRNA of NF-κB p65 in liver were detected by real-time quantitative PCR , and protein expression were detected by Western blotting. Result Compared with the control group , the body weight gain and food efficiency ratio (FER) of experimental group were decreased (P 〈 0.01 ) , but mRNA and protein expression of NF-κB p65 were much higher , and showed very significant difference (P 〈0.01). Conclusion Welfare impaired caused by the stress of experimental procedures , the expression of NF-κB in rats' liver are much higher than before. It shows that NF-κB participate in the process of welfare impaired.
