冠心止痛胶囊含药血清内皮保护作用与体内给药时效关系研究 被引量:1

摘要 目的:观察冠心止痛胶囊含药血清对过氧化氢(H2O2)损伤内皮细胞的保护作用与体内给药的时效关系。方法:建立体外培养的HUVECs细胞H2O2氧化损伤模型,以冠心止痛胶囊不同给药方案所制备的含药血清对氧化损伤内皮细胞存活率、乳酸脱氢酶活性、一氧化氮、内皮素-1、组织型纤溶酶原激活物、组织型纤溶酶原激活物抑制剂的含量为指标,评价不同方案制备的含药血清对内皮细胞的保护作用。结果:不同方案所制备冠心止痛胶囊含药血清对H2O2诱导的HUVECs损伤均具有保护作用,连续3 d、5 d、7 d给药组含药血清的药效明显强于单次给药,连续5 d、7 d组的含药血清作用强于3 d组,但连续5 d、7 d组含药血清药效无统计学差异。结论:冠心止痛胶囊含药血清的药效与给药时间存在一定时效关系,以2次/d连续5 d给药后所制备的含药血清药理效应最佳。 Objective : To study the effect of medicated serum prepared with Guanxin Zhitong capsule in different time -effects on human umbilical vein endothelial cells. Methods:The oxidative injury models induced by H2O2 were established by human umbilical vein endothelial cells and then the cell viability, lactic dehydrogense, nitric oxide, endothelin - 1, tissue plasminogen activator and plasminogen activator inhibitor - 1 were analyzed. They were used to study the time - effect of medicated serum prepared with Guanxin Zhitong capsule, Results:Medicated serum prepared with Guanxin Zhitong capsule had a significantly protective effect on HUVECs injured by H2O2. The 3d group, 5d group and 7d group were better than the single administration group according to protecting HUVECs, while the latter two groups were better than the 3 d group. However, there was no significant difference between 5 d group and 7 d group according to the protective effect on HUVEC injured by H2O2. Conclusion:There is a time - effect relationship of medicated serum prepared with Guanxin Zhitong capsule for the treatment of HUVECs injured by H2O2. Administration with two times per day for five days has the optimal efficacy.
