摘要
目的测定不同产地、不同采收期新疆圆柏叶中穗花杉双黄酮的含量。方法采用高效液相色谱法,以穗花杉双黄酮为指标,色谱条件为Phenomenex ODS-A C_(18)柱(250mm×4.6mm,5μm),流动相为乙腈-0.1%磷酸,梯度洗脱,柱温30℃,流速1.0mL/min,检测波长337nm。结果在0.4~40μg/mL范围内穗花杉双黄酮峰面积与进样浓度呈良好的线性关系,回归方程:Y=43 837 X+6 797.6(r=0.999 9);平均回收率为100.83%,RSD为1.37%;不同产地新疆圆柏叶中穗花杉双黄酮平均含量为1.44mg/g,不同采集时间新疆圆柏叶中穗花杉双黄酮平均含量为1.42mg/g。结论该方法灵敏准确,重现性好,可用于新疆圆柏叶中穗花杉双黄酮的含量测定,为新疆圆柏叶药材的开发利用提供理论依据。
Objective Determination the content of amentoflavone in different areas and different harvest period of Xinjiang Juniperus sabina L. leaves. Methods HPLC method was used with amentoflavone as index, Phenomenex ODS-A C38 column chromatography conditions (250mm×4.6mm,5μm), acetonitrile-0.1% phosphoric acid, gradient elution, the column temperature was 30 degrees, a flow rate of I mL/min, the detection wavelength of 337 nm. Results There are good liner relationships between the peak area and concentration at the range of amentoflavone in the leaves of Juniperus sabina L. were 0.4-40μg/mL, Y=43 837X +6 797.6 (r =0.999 9). The average recovery rate was 100.83% with RSD of 1.37%. The average content of amentoflavone from different habitats of Juniperus sabina L. was 1.44 mg/g, and the average content of amentotaxus collected in different time of Juniperus sabina L. was 1.42 mg/g. Conclusion The method is sensitive and accurate, reproducible and can be used to determine the amentoflavone of Junierus Sabina L. leaves, and to provide scientific basis for exploitation and utilization of Junierus Sabina L. leaves medicinal materials.
