甲状腺癌中CD105及内皮抑素蛋白的表达意义 被引量:6

Significance of expression of Endoglin(CD105) and Endostatin in thyroid carcinoma

摘要 目的检测滤泡性甲状腺癌中Endoglin(又称CD105)和内皮抑素(Endostatin,ES)蛋白表达情况,探讨滤泡性甲状腺癌中血管内皮增殖与肿瘤转移侵袭的相关性。方法收集重庆市第九人民医院及中国医科大学附属第一人民医院甲状腺癌存档蜡块,包括甲状腺乳头状癌(papillary thyroid carcinoma,PTC)130例,滤泡状癌(follicular thyroid carcinoma,FTC)40例,甲状腺未分化癌(undifferentiated thyroid carcinoma,UTC)30例,制作组织芯片,采用免疫组织化学染色法检测滤泡性甲状腺癌中CD105和ES蛋白的表达情况。结果 CD105在PTC、FTC和UTC样本中的表达水平依次递增,平均IRS评分分别为6.1±0.7、7.3±0.8和10.6±1.4(P<0.01);CD105的表达水平与性别、年龄无关,而与癌灶发生甲状腺外侵袭、淋巴结转移和远处转移相关;在随访期内发生甲状腺癌复发或死亡患者的CD105表达水平较病情稳定患者明显升高(P<0.01)。ES在PTC、FTC和UTC样本中的表达水平依次递增,平均IRS评分分别为5.6±2.0、7.0±1.8和9.1±1.2(P<0.01);ES蛋白表达水平与性别、年龄无关,而与腺外侵袭、淋巴结转移或远处转移相关;在随访期内发生甲状腺癌复发或死亡患者的ES表达水平较病情稳定患者明显增高(P<0.01)。同一病例的CD105表达评分与ES表达评分呈正相关(r=0.313,P<0.01)。结论 CD105及ES在不同侵袭性的甲状腺癌组织中的表达存在差异,且具有相关性,可能参与了甲状腺癌的侵袭、转移,导致预后改变,提示其可作为甲状腺癌靶向治疗的靶点之一,为甲状腺癌的治疗提供新的研究方向。 Objective To investigate the expression of Endoglin(CD105) and Endostatin(ES) in follicular thyroid carcinoma as well as the relationship of proliferation of vessel endothelial cells to invasion and metastasis of tumor.Methods The thyroid lesion paraffin blocks of 200 cases from the First Affiliated Hospital of China Medical University and The Ninth People′s Hospital of Chongqing were collected, including papillary thyroid carcinoma(PTC)in 130 cases,follicular carcinoma(FTC) in 40 cases, and undifferentiated thyroid carcinoma(UTC) in 30 cases, which were prepared into tissue microarray and determined for expressions of CD105 and ES by immunohistochemical staining. Results The expression level of CD105 increased in turn in PTC, FTC and UTC samples, of which the mean IRS scores were 6. 1 ±0. 7, 7. 3 ± 0. 8 and 10. 6 ± 1. 4 respectively(P〈0. 01). The expression level of CD105 was unassociated with gender and age, while was associated with the occurrence of thyroid carcinoma invasion, lymph node metastasis and distant metastasis,which was significantly higher in the patients with recurrence of thyroid cancer or the patients dying during the follow-up period than in the patients with stable condition(P〈0. 01). However, the expression level of ES increased in turn in PTC, FTC and UTC samples, of which the mean IRS scores were 5. 6 ± 2. 0, 7. 0 ± 1. 8 and 9. 1 ± 1. 2 respectively(P〈0. 01). The expression level of ES was unassociated with gender and age, which was associated with glandular outside invasion, lymph node metastasis or distant metastasis in thyroid cancer tissues, which was significantly higher in the patients with recurrence of thyroid cancer or dying during the follow-up period than in the patients with stable condition(P〈0. 01). The score for CD105 expression was positively correlated with that for ES expression in the same case(r =0. 3 13, P〈0. 01).
