摘要
为了获取高质量的油梨果肉DNA,以油梨果肉为试材,比较改良SDS法、改良CTAB法和试剂盒法对油梨果肉DNA的提取效果。结果表明:上述3种方法均可从油梨果肉中提取DNA,但改良SDS法与改良CTAB法所获得的DNA浓度都在600μg/m L以上,纯度(OD260nm/OD280nm)均在1.8左右,纯度高且完整性较好,可用于未来分子标记分析。
In order to obtain high-quality DNA from the pulp of avocado, three DNA extraction methods,the modified SDS method, the modified CTAB method, and the commercial plant genomic DNA Kit were conducted to compare their effects of DNA extraction from the pulp of avocado. The results showed that all the three methods were available for DNA extraction from the pulp of avocado, but the DNAs extracted by using the modified SDS method and the modified CTAB method were much higher in concentration(more than 600 μg/m L), purity(OD260nm/OD280 nmwere at about 1.8) and integrity, and could be used for molecular marker analysis in the future.
