骨髓间充质干细胞联合环磷酸腺苷葡胺治疗心力衰竭大鼠的实验研究 被引量:3

摘要 目的探究分析骨髓间充质干细胞(MSC)联合环磷酸腺苷葡胺(MCA)治疗心力衰竭大鼠的实验疗效。方法构建阿奇霉素性心肌病心力衰竭大鼠实验模型30只,采用随机数字表法将实验大鼠分为三组,各10只,分别为MSC组,MCA组,MCA+MSC组,分别给予相应的药物干预。比较不同组别实验大鼠治疗4周后心功能指标水平及血流动力学参数变化情况。结果经4周不同干预治疗后,各组大鼠左室舒张末期内径(LVEDD)、左室收缩末期内径(LVESD)、左室舒张末容积(LVEDV)、左心室收缩末容积(LVESV)等心功能指标水平均较治疗前明显降低(P<0.05),各组左心室射血分数(LVEF%)水平则较治疗前明显增高(P<0.05),且MCA+MSC组大鼠LVEDD、LVESD、LVEDV、LVESV明显低于MSC组和MCA组(P<0.05),其LVEF%水平则明显较MSC组和MCA组偏高(P<0.05)。三组大鼠左室收缩压(LVSP)、左室压力上升最大变化速率(+dp/dtmax)和左室压力下降最大变化速率(-dp/dtmax)较治疗前均明显增高(P<0.05),且MCA+MSC组大鼠增高幅度明显优于MSC组和MCA组大鼠(P<0.05);各组大鼠左室舒张压(LVDP)则明显低于治疗前(P<0.05),且MCA+MSC组大鼠LVDP明显低于MSC组和MCA组大鼠(P<0.05),组间差异具有显著统计学意义。结论环磷酸腺苷葡胺可通过诱导骨髓间充质干细胞分化提高干细胞的移植成功效率,促进心肌特异性蛋白的表达,骨髓间充质干细胞与环磷酸腺苷葡胺联合应用对于治疗心力衰竭疗效显著。 Objective To explore treatment the effect of bone marrow mesenchymal stem cells combined with cyclic adenosine phosphate meglumine for the treatment of heart failure rats.Methods Build 30 azithromycin cardiomyopathy heart failure rat models,using the random number table method experimental rats were divided into three groups,MSC group,MCA group and MCA + MSC group and 10 rats in each group.Cardiac function index level and changes in hemodynamic parameters 4 weeks after treatment were compared.Results After treatment for 4 weeks different intervention,the level of left ventricular end- diastolic diameter(LVEDD),left ventricular end systolic diameter(LVESD),left ventricular end- diastolic volume(LVEDV),left ventricular systolic volume(LVESV) at the end of the cardiac function indexes were significantly lower than those of before treatment(P〈 0.05).The left ventricular ejection fraction(LVEF)%level was significantly higher than that before treatment(P〈 0.05),and LVEDD in MCA + MSC group rats,LVESD,LVEDV,LVESV were significantly lower than those of the MSC and MCA group(P〈 0.05).The level of LVEF%was significantly higher in the MSC group and MCA group(P 〈0.05).The level of left ventricular systolic pressure(LVSP),left ventricular pressure rise rate(+ dp/dtmax) and left ventricular pressure decreases maximum change rate(dp/dtmax) before treatment were significantly higher(P〈 0.05),and those in MCA + MSC group rats increased amplitude was obviously better than MCA + MSC group(P 〈0.05).Left ventricular diastolic pressure(LVDP) was significantly lower than that before treatment(P 〈0.05),and LVDP in MCA + MSC group was obviously lower than that of MSC and MCA group rats(P 〈0.05) with significant difference.Conclusion Cyclic adenosine phosphate meglumine can be improved by inducing bone marrow mesenchymal stem cells to differentiate to improve transplant success efficiency,promote the expression of cardiac muscle specific protei
