热性惊厥大鼠海马组织和皮质神经元中连接蛋白Cx36的表达及意义 被引量:2

Expression and significance of connexin Cx36 in hippocampus and cortical neurons of febrile seizure rats

摘要 目的探讨缝隙连接蛋白Cx36在热性惊厥大鼠海马组织和皮质神经元中的表达。方法将大鼠按随机数字表法分为2组:正常对照组与实验组。实验组根据热水浴法建立惊厥大鼠模型,按大鼠是否惊厥再分为高热对照组和热性惊厥组,采用Westernblot和免疫荧光方法分析正常对照组、高热对照组和热性惊厥组大鼠海马组织和皮质神经元Cx36蛋白表达,多组样本均数比较采用单因素方差分析,均数的两两比较采用LSD法,方差不齐者采用Tamhane’s检验。结果热水浴诱导大鼠惊厥的潜伏期、惊厥持续时间及惊厥时的体温分别为(4.39±0.08)min、(5.38±0.07)min和(41.87±0.06)℃。Westernblot结果表明,热性惊厥组惊厥发作10次后,正常对照组、高热对照组和热性惊厥组3组比较,Cx36蛋白在海马区及皮质区表达呈逐渐降低趋势,热性惊厥组降低程度最明显。诱导惊厥发作1次、5次和10次后,与正常对照组和高热对照组比较,热性惊厥组皮质和海马神经元Cx36蛋白表达均明显降低,且随惊厥次数增加,皮质(0.104±0.012)和CAl(0.091±0.011)、CA3(0.090±0.011)和DG区(0.092±0.012)海马神经元Cx36蛋白表达较正常对照组(0.212±0.017、0.167±0.013、0.159±0.014、0.171±0.013)和高热对照组(0.189±0.006、0.144±0.008、0.129±0.005、0.165±O.011)明显降低,差异均有统计学意义(P均〈0.05)。此外,Cx36表达降低程度与惊厥次数有关。结论随热性惊厥发作次数增加,大鼠海马和皮质神经元中Cx36表达明显下降,可能导致惊厥阈值降低,更易于反复发作。 Objective To explore the potential changes of connexin Cx36 in hippocampus and cortical neurons of rats with hyperthermia - induced convulsion. Methods Rats were divided into 2 groups according to the random number table method:nomlal control group and experimental group. Febrile convulsion model was elicited through im- mersion in warm water. The experimental group was generated following febrile convulsion model:hyperthermia group and febrile convulsion group. Among normal control group, hyperthermia group and febrile convulsion group, western blot analysis and immunofluoreseenee labeling techniques were used to examine the expression of Cx36 protein in the hippo- campus and cortex area. One - Way ANOVA was used to compare the mean of multiple sample ,the LSD test was used to compare the two means. Tamhane's test was used when variance were uneven. Results The incubation period, seizure duration and temperature were (4.39 ± 0.08 ) min, ( 5.38 ± 0.07 ) min, ( 41.87 ± 0.06 ) ~C after hyperthermia-in- duced convulsion ,respectively. Western blot analysis showed that the expression of Cx36 protein in the hippocampus and cortex area decreased gradually after 10 times of seizure in normal control group, hyperthermia group and febrile convulsion group, and the febrile convulsion group decreased most obviously. Compared with normal control group and hyperthermia group,respectively,in febrile convulsion group Cx36 expression obviously decreased in the hippocampus and cortex in rats with 1,5,10 seizure times induced by hyperthermia, and with the increase of number of induced con- vulsion,the expression of Cx36 was significantly decreased in the cortex (0. 104 ± 0. 012) and CA1 (0.091± 0. 011 ),CA3 (0.090±0. 011 ) and DG (0. 092±0. 012) areas of hippocampal neurons compared with the normal control group (0. 212 ± 0. 017,0. 167±0. 013,0. 159±0. 014,0. 171±0. 013 ) and the hyperthermia group (0. 189±0. 006,0. 144±0. 008,0. 129±0.005,0. 165±0.011 ) ( all P 〈 0.05 �
