摘要
以木聚糖为唯一碳源,采用平板筛选和摇瓶发酵相结合的方法,从腐朽木头中分离、筛选到一株高产木聚糖酶菌株XE-13-1根据菌株的形态特征,并结合18S rDNA序列分析,初步鉴定菌株X E-13-1为桔绿木霉(Trichoderma citrinoviride)。经测定,菌株X E-13-1所产木聚糖酶的最适反应温度为50℃,最适反应pH值7,具有较好的pH稳定性。在30℃、pH 5~10的条件下保温2 h,该酶仍具有78%以上的酶活力。Mg^(2+)、Fe^(2+)、K^+、Zn^(2+)对该酶具有明显的促进作用,Cu^(2+)对该酶具有明显的抑制作用。酶学性质研究结果表明,该酶在工业应用上具有较好的前景。
Using xylan as unique only carbon source, a high xylanase-producing strain XE-13-1 was screened from the rotten wood by plate screening and shake-flask fermentation method. According to the strain morphological characteristics and 18S rDNA sequence analysis, the strain XE-13-1 was identified as Trichoderma citrinoviride. The optimal reaction temperature and pH of xylanase produced by strain XE-13-1 were 50 ℃ and 7, respectively. The xylanase had good pH stability. The xylanase activity was more than 78% after preservation for 2 h at 30 ℃ and pH 5-10. Mg2+, Fe2+, K+ and Zn2+ had obvious promoting effect on the xylanase, however, Cu2+ had obvious inhibitory effect on the xylanase. The results of enzymatic properties indicated that the xylanase had good prospect in industrial application.
