摘要
通过透明圈法从土壤中筛选出55株产木聚糖酶的细菌菌株,经产酶发酵培养基复筛及SDS-PAGE酶谱分析得到1株产高分子量木聚糖酶细菌菌株Paenibacillus sp.39631。在单因素实验的基础上,运用响应面分析,对影响产酶发酵的各因素进行优化,得到的最佳产酶发酵条件:碳源(80目玉米芯粉)0.8%,氮源(牛肉膏)1%,初始值pH7.2,培养温度30℃,转速158 r/min,接种量2%,装液量50 mL。在最佳培养条件下,经72 h液态发酵,酶活力达到44.12 U/mL,比优化前提高了4.36倍。
