摘要
甲烷氧化菌是一类能以甲烷和甲醇作为唯一碳源和能源进行生长的微生物。利用甲烷氧化菌选择性培养基从健康奶牛粪便样本中分离、筛选多株甲烷氧化菌,选择其中一株对甲烷代谢率较高的菌株命名为M17,并对其进行形态学观察、16S rDNA同源性比对以及理化性质分析的研究。结果表明,该菌株属于甲烷氧化菌Methylobacterium属,能够利用多种碳源进行生长,但以甲醇为碳源生长最为良好,最适生长温度为35℃,最适pH为6.8左右。
Methanotroph was a kind of bacteria which could use methane as the sole carbon source and energy for their growth. The multiple strains of methanotrophs were isolated and purified using methanotrophs selective media from the stool sample of healthy dairy cattle. One methanotroph was chosen that had high metabolism of methane and named M17. The morphology, 16S rDNA homology alignment and physieochemical property of M17 methanotroph were detected. The results showed that M17 belonged to Methylobacterium,which could use a variety of carbon sources to growth, especially for methanol. The cultivation temperature of M17 was 35 ℃ and pH was 6.8.
