摘要
血浆蛋白广泛地用于临床疾病治疗和作为药物载体,白蛋白、球蛋白和纤维蛋白原是最常用的3种血浆蛋白,其中白蛋白和球蛋白是最有潜力的药物载体。本文主要综述白蛋白作为多肽类药物载体的3种载药方法,包括体外共价融合、非共价键结合及纳米粒包埋。特别阐述了一种新型"白蛋白/结合肽-多肽"的结合方式,在延长药物半衰期的同时保持药物多肽的生理活性。这种新型多肽缓释方法对加速开发多肽药物有促进作用。
Albumin, globulin and fibrinogen are the three most commonly used plasma proteins, among which albumin and globulin are of the most potential for the treatment of clinical disease.This review mainly focuses on three kinds of methods which use albumin as a carrier for peptide drug delivery, including in vitro covalent fusion, noncovalent bonding and nanoparticle embedding.In particular, a novel delivery strategy based on the interaction of albumin with albumin-binding peptide linked to a therapeutical peptide for extending the half-life of the therapeutical peptide is discussed.This new method will facilitate the clinical application of peptides.
