摘要
采用喷雾干燥法制备葡萄籽原花青素微囊。以微囊成型情况、对指标成分原花青素B1和原花青素B2的包封率为考察指标,采用单因素试验考察进料速度、进风温度、固形物含量、壁材组成、壁芯比对微囊的影响。所得优化工艺和处方为:进料速度4 ml/min,进风温度80℃,固形物含量25%,壁材组成为玉米肽和阿拉伯胶3∶1;壁芯比为1.5∶1。验证试验表明,3批优化微囊对原花青素B_1和原花青素B_2的包封率为80.89%和84.64%,粒径(1.34±0.12)μm。显微观察显示微囊表面光滑,无黏连。体外释放试验表明,该微囊中原花青素B_1和原花青素B_2的0.5和12 h累积释放率均约为26%和95%,提示制品具有一定的缓释作用。
Grape seed procyanidin microcapsules were prepared by spray drying method. The effects of feed rate, inlet air temperature, solid content, wall materials, ratio of wall materials to core materials on the formation and encapsulation efficiencies for procyanidin B1 and B2 of the microcapsules were investigated by single factor experiment. The optimal process and formulation parameters were as follows: feed rate was 4 ml/min, inlet air temperature was 80 ℃, solid content was 25%, the wall materials were the mixture of corn peptide and arabic gum in a weight ratio of 3∶ 1, and the ratio of wall materials to core materials was 1.5∶1. The results of validation test showed that the average encapsulation efficiencies for procyanidin B1 and B2 of three batches of the optimal microcapsules with the particle size of (1.34±0.12)μm and smooth surfaces were 80.89% and 84.64%. The microscopic observation also showed that there was no adhesion between the microcapsules. The in vitro release of procyanidin B1 and B2 from the microcapsules at 0.5 and 12 h were about 26% and 95%, which indicated that the product had sustained-release characteristics.
