摘要
通过西瓜抗病和感病样品不同比例的混合,研究不同混样比例对西瓜纯度d Caps分子标记鉴定的影响。结果表明,随着感病样品混合比例的降低,116 bp感病识别位点的电泳条带清晰度逐渐降低,在混合比例低于1∶10时仍可以识别出清晰的杂株位点,推荐混样株数为10株,在该数量株数的混合下,样品中只要出现1个杂株位点就可以被检测出来,采用该混合方式能够大幅度缩短西瓜纯度的分子标记鉴定工作量。
Experiments on the effect of different proportion of mixture ratio on the dCaps molecular marker identification of watermelon was conducted by mixture of watermelon disease resistant and susceptible samples, the results showed that the resolution of the electrophoresis bands of the 116 bp recognition site was gradually decreased with the decrease of the proportion of the mixture ratio of the susceptible samples, the heterozygous loci could be identified still when the mixed ratio is lower than 1 : 10, therefore, 10 plants was recommend as mixed number of samples.
