抗猪嗜血支原体中药单体的体外筛选 被引量:3

Screening natural compounds with anti-M.suis activity in vitro

摘要 为体外筛选具有杀灭猪嗜血支原体(M.suis)作用的中药单体,应用血液直接PCR扩增中药单体作用不同时间后培养体系中的M.suis,通过比较电泳条带灰度值筛选出具有抗M.suis作用的中药单体;应用qPCR测定所筛选中药单体作用后培养体系中M.suis的拷贝数,通过杀灭率来评估中药单体对M.suis的杀灭效果。结果筛选出丹参酮ⅡA磺酸钠、甘草酸二钾、秦皮甲素、黄芩苷和肉桂酸5种具有体外抗M.suis作用的中药单体,其中在药物作用18h,丹参酮ⅡA磺酸钠、甘草酸二钾和秦皮甲素对M.suis杀灭率分别为76.9%,90.6%和79.2%。体外试验表明,丹参酮ⅡA磺酸钠、甘草酸二钾和秦皮甲素对M.suis的杀灭效果较好,是理想的抗M.suis目标药物,具有潜在的临床应用与研究价值。 In order to screening natural compounds which are able to kill Mycomplasma suis in vitro,natural compounds were added to positive blood infected M. suis and blood direct PCR method was applied to amplify the specific gene segments of M. suis at different times. The gray values of electrophoretic bands were used to preliminarily assess the anti-M, suis activity of tested natural compounds. The qPCR method was used to detect the M. suis copy numbers of positive blood that interacted with valid compounds at different times and the killing ratio was calculated to evaluate the anti-M, suis activity of candidates. The results indicated that five anti-M, suis natural compounds were preliminarily screened in vitro,including dipotassium glycyrrhizinate, sodium tan- shinone IIA sulfonate,aesculin,baicalin and cinnamic acid. At 18 hours, the killing ratio of sodium tanshinone IIA sulfonate,dipotassium glycyrrhizinate and aesculin had reached at 76.9%, 90.6% and 79.2% ,respectively. In conclution, the killing ratioes of sodium tanshinone IIA sulfonate, di- potassium glycyrrhizinate and aesculin had reached above 70.0% at 18 hours and they might be the ideal anti-M, suis drugs and have a potential clinical application and research value.
