摘要
目的探讨脂联素(APN)抑制氧化型低密度脂蛋白(ox-LDL)诱导的血管平滑肌细胞(VSMCs)泡沫化是否与干扰Toll样受体4(TLR4)及其介导的炎症反应相关。方法组织贴块法培养C57BL/6J背景野生型及TLR4基因敲除(TLR4-/-)小鼠主动脉VSMCs。油红O染色观察VSMCs内脂质聚积;酶法检测VSMCs内胆固醇水平;Western blot检测VSMCs TLR4蛋白表达;ELISA方法检测VSMCs培养液中炎症因子IL-6和TNF-α的表达。结果 ox-LDL刺激VSMCs 48h显著诱导细胞泡沫化;ox-LDL诱导VSMCs泡沫化过程中伴随着TLR4及其下游炎症因子IL-6、TNF-α表达的上调;而对于TLR4-/-型VSMCs,ox-LDL刺激并不能有效诱导VSMCs泡沫化及IL-6、TNF-α表达上调;APN显著抑制ox-LDL诱导的VSMCs泡沫化,伴随着TLR4及其下游炎症因子IL-6、TNF-α表达的下调。结论 APN可通过干扰TLR4介导的炎症反应抑制ox-LDL诱导的VSMCs泡沫化。
Objective To investigate whether adiponectin(APN)inhibiting oxidized low density lipoprotein(ox-LDL)induced foam cell formation of vascular smooth muscle cells( VSMCs) is associated to interfere toll like recepter 4 (TLR4)and its mediated inflammation reactions. Methods Primary VSMCs were isolated from the thoracic aorta of the male(C57BL/6J)wild-type mice and TLR4 knockout(TLR4-/- )mice. Oil red O staining was used to observe the intracellular lipid deposition in VSMCs; enzymatic assay was used to analyze the intracellular total cholesterol content in VSMCs ; the expression of TLR4 in VSMCs was detected by Western blot;the levels of IL-6 and TNF-a in the VSNCs culture solution were quantified by using ELISA. Results Ox-LDL stim- ulating VSMC for 48 h significantly induced its foam cell formation, and this process was accompanied by the up-regulation of TLR4,downstream inflammatory factors IL-6 and TNF-α expressions however, in TLR4 -/- VSMCs, the ox-LDL stimulation failed to effectively induce the VSMC foam cell formation,as well as the expression up-regulation of IL-6 and TNF-α;APN signifi- cantly inhibited the ox-LDL induced-VSMC foam cell formation, and down-regulated the expressions of TLR4, IL-6 and TNF-α. Conclusion APN could inhibit ox-LDL induced VSMC foam cell formation by interfering TLR4 mediated inflammation reaction.
