摘要
目的探讨褪黑素(MT)对动脉粥样硬化(AS)模型兔血脂、主动脉Toll样受体4(TLR4)蛋白表达、血炎症因子水平和AS进展的影响。方法复制实验性兔AS模型。36只新西兰大白兔,随机均分为3组。正常组:给予普通饲料喂养;模型组:给予高胆固醇饲料喂养;MT组:给予高胆固醇饲料加MT喂养。12周后分别检测血清血脂[总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、三酰甘油(TG)]水平和血清炎症因子[肿瘤坏死因子α(TNF-α)、高敏C反应蛋白(hs-CRP)]水平,观察主动脉病理形态学变化,免疫组织化学法和Western blot法检测兔主动脉TLR4蛋白水平。结果 12周高脂喂养成功建立实验性兔AS模型;与正常组相比,模型组TC、LDL-C、HDL-C、TG、TNF-α、hs-CRP明显升高,内膜厚度与内膜/中膜厚度比明显升高,动脉TLR4蛋白表达较正常组显著升高(P<0.05)。与模型组相比,MT组血清TC、LDL-C、TG、TNF-α、hs-CRP明显下降,HDL-C明显升高,主动脉中TLR4表达减少,内膜厚度、内膜/中膜厚度比明显降低(P<0.05)。结论MT可能通过改善血脂、降低动脉TLR4蛋白表达和血炎症因子水平抑制实验性兔AS进展。
Objective To investigate the influence of melatonin on the expression of toll-like receptor 4(TLR4),blood lipids,inflammatory cytokines and the development of experimental atherosclerosis in rabbits.Methods 36 New Zealand white rabbits were randomly assigned to three groups,including control group,model group,and melatonin(MT) group.The control group was given ordinary forage,the model group was given high-fat food based on ordinary forage,and the MT group was given MT daily on the basis of food in-take like high-fat group.Serum total cholesterol(TC),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C),triglyceride(TG),tumor necrosis factor-α(TNF-α),and high-sensitivity C-reactive protein(hs-CRP) were detected on the twelfth week of experiment.The structure of arteries was observed.TLR4 expression was detected by immunohistochemistry and Western blot.Results The atherosclerosis model was established successfully.Serum TC,LDL-C,HDL-C,TG,TNF-α,hs-CRP and arterial TLR4 levels as well as the arterial intima and intima-to-media thickness ratio of the model group were significantly higher than those of control group(P0.05).Serum TC,LDL-C,HDL-C,TG,TNF-α,hs-CRP and arterial TLR4 levels as well as the arterial intima and intima-to-media thickness ratio of the MT group were significantly lower than those of model group(P0.05).Conclusion Melatonin might suppress the development of experimental atherosclerosis in rabbits by decreasing TLR4 protein and inflammatory cytokines
