摘要
microRNA(miRNA)在不同物种间进化具有高度保守性,且在动物生长发育过程中有重要作用。为研究miR-92的进化历史、miR-92在不同组织的表达差异性和在生长发育过程中的作用,本文运用多序列比对分析序列相似性,并构建系统进化树分析miR-92基因的进化特点,运用SYBR Green System PCR技术构建了miR-92在鸡不同发育阶段11个组织中的表达谱,同时用TargetScan 7.0与PicTar两种不同生物学软件对其进行靶基因预测,两个软件靶基因的交集分别进行GO和KEGG富集分析。结果表明:(1)miR-92的进化方向与物种从低等到高等的进化趋势相一致,在不同物种中的基因位置和序列上都表现出较高的相似性,这与miRNA在物种间高度进化保守性的特点基本一致;(2)同一个时期,不同组织部位的表达有组织特异性,在1日龄的鸡中,肾脏,腿肌和心脏有较高的表达量。在成年鸡中,表达量较高的组织为肺脏,胸腺和下丘脑。在同一组织的1日龄和成年鸡中的表达呈现时序性,除脾脏的表达量差异不显著外,其他10个组织miR-92的表达量差异显著(P<0.05);(3)两个软件预测到的交集靶基因有157个,GO富集结果显示靶基因主要参与转录过程,RNA代谢过程,DNA结合,转录因子活性,MAPK磷酸酶活性等。显著富集的KEGG通路为MAPK信号通路(P<0.05)。这些结果为进一步研究miR-92基因在机体中所扮演的角色奠定了基础。
microRNA(miRNA)shows highly evolved conservation among different species,and plays an important role in animal growth and development.To investigate the evolutionary history,the expression difference and the function of miR-92 in animal growth and development,multiple sequence alignment was used to construct the evolution tree to analyze the evolutionary features of miR-92 gene,and SYBR Green System PCR was applied to construct different expressions of miR-92 in 11chicken tissues of different growth stages.In the meantime,TargetScan 7.0and PicTar were used to predict target genes of miR-92 and the intersection of the results was analyzed by GO and KEGG pathway enrichment analysis.The results indicate that(1)miR-92 evolution followed the evolutionary trend of the lower animals to the higher animals,with higher similarities in both gene loci and sequence,which was consist with the fact that miRNAs had highly evolutionary conservation.(2)Expressions of different tissues at the same period had tissue specificity.miR-92 was abundant in kindey,heart and crura muscle of 1day-old chicken and was abundant in lung,thymus gland and the hypothalamus of adult chicken.The expressions of miR-92 of same tissue of 1-day-old chicken and adult chicken all present time sequence,apart from insignificant difference for spleen,the miR-92 expression in other 10 tissues were all significantly different(P<0.05).(3)the 157 target genes in the intersection from algorithms PicTar and TargetScan are interference with KEGG pathway and GO analysis.The GO category indicated that these target genes mainly participate in the process of transcription,RNA metabolism,including DNA bonding,transcription factor activity,the activity of MAPK phosphatase and so on.KEGG pathway existed in the pathway of MAPK signaling pathway etc.These results would provide a beneficial reference for further study of miR-92 on the regulation in animal growth and development.
