慢性砷染毒对大鼠附睾上皮细胞凋亡的影响 被引量:5

摘要 目的探讨慢性砷染毒对大鼠附睾上皮细胞凋亡的影响。方法将40只健康清洁雄性SD大鼠随机分为4组,分别为对照(蒸馏水)组和2.4、12.0、60.0 mg/L亚砷酸钠染毒组,每组10只。采用自由饮用方式进行连续染毒6个月。采用TUNEL细胞凋亡染色法测定大鼠附睾上皮凋亡细胞的凋亡情况,采用Western blot法测定附睾组织内caspase-3、caspase-9蛋白的表达水平,并测定精子顶体完整率和精子畸形率。结果与对照组比较,各浓度砷染毒组大鼠附睾管的细胞凋亡率较高,差异均有统计学意义(P<0.05);且随着砷染毒浓度的升高,大鼠附睾管的细胞凋亡程度加重。与对照组比较,各浓度砷染毒组大鼠附睾内caspase-3、caspase-9蛋白表达水平均较高,差异有统计学意义(P<0.05);且随着砷染毒浓度的升高,大鼠附睾内caspase-3、caspase-9蛋白的表达水平呈上升趋势。与对照组比较,12.0、60.0 mg/L砷染毒组大鼠精子顶体完整率均较低,而精子畸形率均较高,差异均有统计学意义(P<0.05);且随着砷染毒浓度的升高,大鼠精子顶体完整率呈下降趋势,而精子畸形率呈上升趋势。结论慢性砷染毒通过激活caspase-3或caspase-9等途径实现大鼠附睾组织的损伤及凋亡。 Objective To explore the effect of chronic arsenic exposure on the apopotosis in epididymal epithelial cells of rats. Methods Forty healthy clean grade male SD rats were randomly divided into four groups,respectively for the control group (distilled water),2.4,12.0,60.0 mg/L sodium arsenite groups, 10 in each group. The rats were exposed to sodium arsenite by free drinking for six consecutive months. The apoptosis rates of the epididymis epithelial cells were deteiminated by TUNEL staining method,the protein expression levels of caspase-3 and caspase-9 in epididymis tissues of rats were detected by Western blot assay, the sperm acrosome intact rate and sperm deformity rate were determinated. Results Compared with the control group,the apoptosis rates of epididymal tubes in each exposure group were higher,the differences were statistically significant (P〈0.05); And with the increase of arsenic concentrations,the apoptosis rates of epididymal tube were increased. Compared with the control group, the levels of caspase-3 and caspase-9 protein expression were higher in each sodium arsenite exposure groups,the differences were statistically significant (P〈0.05); And with the increase of concentration of arsenic poisoning,the levels of caspase-3 and caspase-9 protein expression in epididymis increased. Compared with the control group, the sperm acrosome rates in 12.0,60.0 mg/L sodium arsenite exposure groups were lower,and the sperm malformation rates were higher,the differences were statistically significant (P〈0.05); And with the increase of arsenic concentrations,the rates of sperm acrosome intact were decreased,while the rates of sperm deformity were increased. Conclusion Chronic arsenic exposure can lead to the epididymis tissue injury and apoptosis by activating easpase-3 or caspase-9.
