甘草降低山豆根致小鼠肝毒性的实验研究 被引量:8

Study of Radix Glycyrrhiza on Reducing Hepatic Toxicity Induced by Radix et Rhizoma Sophorae Tonkinensis in Mice

摘要 目的:研究山豆根配伍甘草对山豆根所致实验小鼠肝损害的影响。方法:将50只SPF级昆明小鼠分为山豆根组、山豆根甘草(1∶1)配伍组、山豆根甘草(1∶2)配伍组、山豆根甘草(2∶1)配伍组和空白组。各试验组均给药21d后,观察和比较各组小鼠的生理、生化和病理指标。结果:山豆根组小鼠出现肝损害,小鼠肝脏指数以及血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总胆红素(TBIL)均较空白组升高(P<0.05或P<0.01);甘草不同比例配伍山豆根能减轻山豆根所致实验小鼠的肝脏指数、降低小鼠血清中ALT、AST的增长水平,与山豆根组比较,差异有统计学意义(P<0.01),并能改善实验小鼠的肝脏组织病变。结论:山豆根对肝脏的损害属于对肝细胞的直接毒性作用;甘草能明显减轻山豆根造成的肝损害且以山豆根与甘草1∶2的配伍比例减毒效果较好。 Objective : To study the impacts of the combination of Radix et Rhizoma Sophorae Tonkinensis (RRST) with radix glycyrrhiza on experimental mice hepatic toxicity induced by RRST. Methods : 50 SPF - class Kumning mice were divided into RRST group, the combination of RRST and radix glycyrrhiza (1:1) group,the combination of RRST and radix glycyrrhiza (1: 2 ) group,the combination of RRST and radixglycyrrhiza (2: 1) group and control group. In all of the trial groups after medication for 21 days separately, the indexes of physiology, physiochemistry and pathology of mice were observed and compared among groups. Results: In RRST groups afte rmedication, hepatic toxicity happened in mice. Hepatic indexes, serum ALT, AST and TBIL of mice all increased as compared with control group (P〈0. 05 orP〈0.01). The combination of radix glycyrrhiza at different ratios reduced the increase of hepatic indexes, ALT and AST in experimental mice induced by RRST. Compared with RRST group, the statistical significant differences were obtained (P 〈0.01)and the hepatic tissue disorders in experimental mice were relieved. Conclusion:Hepatic toxicity induced by RRST brings direct toxicity to hepatic cell. Radix glycyrrhiza can alleviate obviously hepatic toxicity induced by RRST and the combination of RRST and radix glycyrrhiza at the ratio of 1 :2 achieves the better efficacy on it.
